Abstract

Pencemaran crude oil dapat menurunkan kualitas lingkungan. N-heksadekana (HXD) adalah kandungan senyawa alifatik hidrokarbon rantai panjang terbanyak dalam crude oil. Kelompok yeast mampu mendegradasi HXD menggunakan enzim sebagai sumber karbon untuk pertumbuhannya. Pada penelitian sebelumnya (2019), isolat yeast W.7.3 dan TPSL cukup potensial mendegradasi crude oil. Alkana hidroksilase (AHs) merupakan enzim kunci dalam degradasi awal HXD sebagai katalis hidroksilasi gugus metil HXD dengan kofaktor nikotinamida adenine dinukleotida hidrogen (NADH). Tujuan penelitian adalah mengetahui perbedaan aktivitas crude AHs dalam mendegradasi HXD oleh strain isolat yeast W.7.3 dan TPSL. Kurva pertumbuhan isolat diamati selama dua kali aklimatisasi pada media Mineral Salt Medium Broth (MSMB) yang ditambahkan 1% HXD + 5% glukosa, dan 1% HXD + 2% glukosa. Crude enzim intraseluler diekstraksi dengan sonikasi setiap hari selama 7 hari inkubasi pada MSMB yang ditambahkan 1 % HXD + 0,1% glukosa. Aktivitas enzim diukur dengan spektrofotometer pada panjang gelombang 340 nm selama 30 menit. Hasil dari penelitian menunjukkan bahwa isolat W.7.3 memiliki aktivitas crude AHs lebih baik dibanding isolate TPSL. Isolat W.7.3 mempunyai aktivitas crude AHs tertinggi pada hari ke- 6 sebesar 821,429 U/mL, sedangkan isolat TPSL pada hari ke-5 sebesar 383,5 U/mL. Tidak ada pengaruh yang signifikan antara isolat yeast W.7.3 dan TPSL terhadap aktivitas crude AHs dalam mendegradasi HXD (p value > 0,05). Berdasarkan data FTIR di hari ke-7 inkubasi oleh isolat W.7.3 dan TPSL terdapat peregangan rantai C-H, C=O, dan C-C oleh AHs di rantai karbon HXD.

Crude oil pollution can reduce environmental quality. N-hexadecane (HXD) is the most abundant aliphatic long-chain hydrocarbon compound in crude oil. Yeast groups are able to degrade HXD using enzymes as a carbon source for growth. In previous research (2019), isolates W.7.3 and TPSL are quite potential to degrade crude oil. Alkane hydroxylase (AHs) is a key enzyme in the initial degradation of HXD as a catalyst for hydroxylation of HXD methyl groups with the cofactor nicotinamide adenine dinucleotide hydrogen (NADH). The aim of the study was to determine the difference in the activity of crude AHs in degrading HXD by yeast isolate strains W.7.3 and TPSL. The growth curves of the isolates were observed during two acclimatizations on Mineral Salt Medium Broth (MSMB) media supplemented with 1% HXD + 5% glucose, and 1% HXD + 2% glucose. Crude intracellular enzyme was extracted by sonication every day during 7 days of incubation on MSMB supplemented with 1% HXD + 0.1% glucose. Enzyme activity was measured by spectrophotometer at 340 nm wavelength for 30 min. The results showed that isolate W.7.3 had better crude AHs activity than isolate TPSL. Isolate W.7.3 had the highest crude AHs activity on day 6 at 821.429 U/mL, while isolate TPSL on day 5 at 383.5 U/mL. There was no significant effect between W.7.3 and TPSL yeast isolates on the activity of crude AHs in degrading HXD. Based on FTIR data on the 7th day of incubation by isolates W.7.3 and TPSL, there is stretching of C-H, C=O, and C-C chains by AHs in the HXD carbon chain
