Abstract

Curculigo latifolia merupakan spesies endemik Pulau Kalimantan dimana memiliki potensi pemanfaatan sebagai obat ataupun pemanis buatan. Namun, spesies ini hanya terdapat di alam liar dan sulit dibudidayakan karena memiliki viabilitas yang rendah. Oleh karena itu digunakan teknik in-vitro dengan rekayasa lingkungan berupa cahaya untuk meningkatkan produktivitas tanaman. Tanaman menyerap jenis cahaya yang disebut Photosynthetically Active Radiation (PAR) dengan menggunakan berbagai jenis fotoreseptor. Sinyal cahaya ini mempengaruhi ekspresi gen dan dapat menstimulasi respons fisiologis salah satunya fotosintesis. Salah satu tahap fotosintesis adalah siklus calvin, dimana terjadi proses fiksasi CO2 yang dikatalis oleh enzim ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco). Rubisco tersusun atas delapan small subunits (SSU) yang dikode oleh multigene family (rbcS) pada nukleus, dan delapan large subunits (LSU), yang dikode oleh single gene (rbcL) di kloroplas. Setiap LSU mengandung satu sisi aktif yang berfungsi mengikat RuBP dan memulai fiksasi karbon. Peningkatan sintesis Rubisco mengakibatkan peningkatan kapasitas fotosintesis. Oleh karena itu penelitian ini bertujuan untuk mengatahui pengaruh jenis cahaya terhadap ekspresi gen rbcL pada lemba (Curculigo latifolia). Penelitian ini menggunakan planlet yang diinduksi dari eksplan biji yang ditumbuhkan pada media MS0 (Murashige and Skoog) dengan perlakuan cahaya LED merah, LED biru, LED putih, serta fluorescence putih. Parameter yang diukur adalah ekspresi gen rbcL. Ekspresi gen rbcL dianalisis secara deskriptif kuantitaif menggunakan Real-Time Quantitative PCR. Hasil penelitian dengan perlakuan paparan jenis cahaya yang berbeda menunjukkan pengaruh terhadap ekspresi gen rbcL pada planlet Lemba (Curculigo latifolia) secara in-vitro. Nilai relatif ekspresi gen rbcL tertinggi terdapat pada planlet dengan perlakuan paparan cahaya biru LED yaitu sebesar 3,81 fold. Sedangkan ekspresi gen relatif terendah terdapat pada planlet yang terpapar cahaya merah LED yaitu sebesar 0,37 fold.

Curculigo latifolia is an endemic species of Borneo Island which has great potential as a medicine or artificial sweetener. However, this species only exists in the wild and is not cultivated. Therefore, in-vitro techniques by controlled environment condition such as light to increase plant productivity. Plants absorb a type of light called Photosynthetically Active Radiation (PAR) using different types of photoreceptors. This light signal affects gene expression and can stimulate physiological responses, include photosynthesis. One of the stages of photosynthesis is the Calvin cycle, where the process of CO2 fixation catalyzed by the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) occurs. Rubisco is composed of eight small subunits (SSU) encoded by a multigene family (rbcS) in the nucleus, and eight large subunits (LSU), encoded by a single gene (rbcL) in chloroplasts. Each LSU contains one active side that functions to bind RuBP and initiate carbon fixation. Increased Rubisco synthesis results in increased photosynthetic capacity. Therefore, this study aims to determine the effect of light type on rbcL gene expression in lemba (Curculigo latifolia). This study used planlets induced from seed explants grown on MS0 (Murashige and Skoog) media with light treatment of red LED, blue LED, white LED, and white fluorescence. Parameters rbcL gene expression were analyzed descriptively quantitatively using Real-Time Quantitative PCR. The results showed that the treatment of exposure to different types of light had an effect on the expression of the rbcL gene in Lemba (Curculigo latifolia) planlets in-vitro. The highest relative value of rbcL gene expression was found in planlets with blue LED light exposure treatment which amounted to 3.81 fold. While the lowest relative gene expression was found in planlets exposed to LED red light which amounted to 0.37 fold.
