Abstract

Konsumsi biota laut yang mengandung logam berat akan terakumulasi dan berdampak pada komposisi mikrobioma pada saluran pencernaan manusia yang berperan penting dalam regulasi sistem di dalam tubuh. Beberapa filum dari mikrobioma usus dapat bertahan karena memiliki beragam mekanisme resistensi untuk menangkal cekaman logam berat karena adanya beragam gen resisten dan sistem ketahanan. Namun, penelitian mengenai korelasi kadar logam berat terhadap gen resisten pada mikrobioma usus masih belum dilakukan. Studi efek pencemaran logam berat pada gen resisten mikrobioma sangat mendukung wawasan mengenai pemulihan lingkungan paparan logam berat. Oleh karena itu, penelitian ini bertujuan untuk menginvestigasi kadar logam berat pada usus manusia, menganalisis korelasi antara logam berat dengan komposisi mikrobioma usus manusia, dan kelimpahan relatif gen resisten logam berat pada mikrobioma usus manusia. Sampel feses diperoleh dari 20 partisipan sehat dari 2 populasi penelitian yaitu populasi dari pesisir yang terpapar kontaminasi logam berat tinggi di Kenjeran, Surabaya, Indonesia (7 pria dan 2 wanita) dan populasi dari dataran tinggi yang terpapar kontaminasi logam berat rendah di Pacet, Mojokerto, Indonesia (5 pria dan 6 wanita) kemudian dilakukan ekstraksi DNA, pengukuran kemurnian DNA dan analisis Metagenomic Next-Generation Sequencing lalu dilakukan pengukuran kadar logam berat dengan X-Ray Fluorescence dan pengabuan. Analisis dilakukan secara statistik yaitu uji normalitas Shapiro Wilks, uji korelasi Pearson dan Spearman Rank. Logam berat yang ditemukan pada feses manusia di Kenjeran dan Pacet meliputi Fe, Mn, Cu, dan Zn. Korelasi yang ditemukan antara logam berat Mn, Fe, dan Cu dengan kelimpahan Prevotella copri bernilai negatif. Sedangkan korelasi yang ditemukan antara logam berat Mn dengan Collinsella, Clostridiaceae, Coriobacteriaceae bernilai positif

Consumption of marine biota containing heavy metals will accumulate and have an impact on the composition of microbes in the human digestive tract which play an important role in the regulation of systems in the body. Several phyla of the gut microbiota survive because they have various resistance mechanisms to heavy metal stress due to the presence of various resistance genes and resistance systems. However, research on the correlation of heavy metal levels to resistance genes in the gut microbiota has not been carried out. The study of correlation between heavy metal pollution and microbiota resistance genes strongly supports insights regarding environmental restoration of heavy metal exposure. Therefore, this study aims to investigate the levels of heavy metals in the human intestine, analyze the correlation between heavy metals and the composition of the human gut microbiota, and the relative abundance of heavy metal resistance genes in the human gut microbiota. Feces samples were obtained from 22 healthy participants from 2 study populations, population from the coast exposed to high heavy metal contamination in Kenjeran, Surabaya, Indonesia (9 men and 2 women) and population from the highland exposed to low heavy metal contamination in Pacet, Mojokerto, Indonesia (5 men and 6 women). Next, DNA extraction and DNA quantification are needed in the analysis using Metagenomic Next-Generation Sequencing, and then heavy metal levels are measured with X-Ray Fluorescence and ashing. The result is analyzed by using the Shapiro-Wilks normality test and the Pearson and Spearman Rank correlation tests. Heavy metals found in human feces in Kenjeran and Pacet include Fe, Mn, Cu and Zn. The correlation found between the heavy metals Mn, Fe, and Cu and the abundance of Prevotella copri was negative. While the correlation found between the heavy metal Mn and Collinsella, Clostridiaceae, Coriobacteriaceae is positive.
