Abstract

Infeksi penyakit hewan komoditas industri perikanan semakin meningkat. Vibrio spp. bertanggung jawab atas infeksi penyakit Acute Hepatopancreatic Necrosis Disease (AHPND) yang dapat berkembang dengan cepat dan menyebabkan mortalitas hingga 100% dalam 20-30 hari. Penelitian ini dilakukan untuk mengetahui potensial mikroba yang diisolasi dari ekosistem perairan tambak udang yang memiliki sifat antagonis terhadap Vibrio spp. Penelitian dilakukan di Laboratorium Genomik, Badan Riset dan Inovasi Nasional (BRIN), Cibinong, Bogor. Isolasi mikroba dilakukan dengan menggunakan media Lactobacillus de Mann Rogosa Sharpe Agar (MRSA). Purifikasi dan skrining isolat dilakukan berdasarkan karakteristik makroskopis dan mikroskopis. Sebanyak 5 isolat mikroba terpilih dilakukan uji antagonis terhadap Vibrio spp. dengan metode zona bening pada media Mueller-Hinton Agar (MHA). Berdasarkan hasil uji antagonis, diketahui hanya isolat 3.2 yang menunjukkan adanya zona bening di sekitar disc. Uji Minimum Inhibitory Concentration (MIC) dilakukan juga untuk mengetahui konsentrasi terendah yang mampu menghambat bakteri Vibrio spp. Berdasarkan hasil uji MIC, hanya isolat 3.2 yang mampu menghambat Vibrio spp. pada konsentrasi terendah yaitu 0,16 mg/ml. Identifikasi genus dilakukan dengan uji biokimia dan disesuaikan dengan Bergey’s Manual for Determinative Bacteriology. Berdasarkan karakteristik morfologi dan hasil uji biokimia, diketahui isolat 3.2 memiliki karakter kunci yang sama dengan bakteri pada genus Bacillus.

Infection with animal diseases in the fishery industry is increasing. Vibrio spp. responsible for infection with Acute Hepatopancreatic Necrosis Disease (AHPND) which can develop rapidly and cause mortality of up to 100% in 20-30 days. This research was conducted to determine the potential of microbes isolated from aquatic ecosystems of shrimp ponds that have antagonistic properties to Vibrio spp. The research was conducted at the Genomics Laboratory, Badan Riset dan Inovasi Nasional (BRIN), Cibinong, Bogor. Microbial isolation was carried out using Lactobacillus de Mann Rogosa Sharpe Agar (MRSA) media. Purification and screening of isolates was carried out based on macroscopic and microscopic characteristics. A total of 5 selected microbial isolates were subjected to antagonistic assay against Vibrio spp. using the clear zone method on Mueller-Hinton Agar (MHA) media. Based on the results of the antagonist assay, it was found that only isolate 3.2 showed a clear zone around the disc. The Minimum Inhibitory Concentration (MIC) assay was also carried out to determine the lowest concentration capable of inhibiting Vibrio spp. Based on the MIC assay results, only isolate 3.2 was able to inhibit Vibrio spp. at the lowest concentration of 0.16 mg/ml. Genus identification was carried out by biochemical tests and adapted to Bergey's Manual for Determinative Bacteriology. Based on the morphological characteristics and the results of biochemical tests, it is known that isolate 3.2 has the same key characters as bacteria in the genus Bacillus.
