Abstract
Bangunan candi yang terbuat dari batu bata identik dengan permasalahan pelapukan baik secara fisik, kimia, maupun biologi. Faktor biologi penyebab pelapukan batu bata yaitu mikroorganisme, salah satunya berasal dari eksudat yang dikeluarkannya. Bakteri adalah salah satu mikroorganisme yang di dalam habitatnya terdiri dari beberapa spesies yang mempunyai sifat morfologi maupun fisiologis yang berbeda, sehingga membentuk suatu keanekaragaman.Terdapat beberapa metode dalam mempelajari keanekaragaman bakteri, salah satunya menggunakan pendekatan taksonomi numerik fenetik. Tujuan penelitian ini adalah mengetahui keanekaragaman bakteri pada permukaan batu bata candi dengan pendekatan taksonomi numerik fenetik. Isolasi bakteri dilakukan secara aseptik, dilabel, dan dibawa ke laboratorium untuk dikarakterisasi secara morfologi, uji biokimia, dan reaksi fisiologis. Selanjutnya, diuji potensi enzim dan resistensinya terhadap logam berat Pb, Cd, dan Fe. Konstruksi dendogram menggunakan software MVSP (Multi-Variate Statistical Package) dengan pengelompokkan isolat berdasarkan algoritma Unweighted Pair Group Method Averages (UPGMA). Kesamaan antar isolat menggunakan nilai similaritas Simple Matching Coefficient (SSM). Hasil konstruksi dendogram didapatkan 3 cluster yaitu A (4 isolat), B (1 isolat), dan C (2 isolat). Rentang indeks similaritas antar isolat adalah 0,543 - 0,857. Cluster A, B, dan C mempunyai indeks similaritas ≤0,70 menunjukkan bahwa masing-masing anggota cluster adalah spesies yang berbeda. Hasil profile matching karakter kunci ketujuh isolat bakteri teridentifikasi sebagai genus Bacillus (Cluster A), Corynebacterium (Cluster B), dan Mycobacterium (Cluster C).

Temple buildings made of bricks are identical to the problem of weathering physically, chemically, and biologically. Biological factors that cause weathering of bricks are microorganisms, one of which comes from the exudates they release. Bacteria are microorganisms that in their habitat consist of several species that have different morphological and physiological properties, thus forming a diversity. There are several methods for studying bacterial diversity, one of which uses a phenetic numerical taxonomy approach. The purpose of this study was to determine the diversity of bacteria on the surface of temple bricks with a phenetic numerical taxonomy approach. Bacterial isolation was done aseptically, labeled, and brought to the laboratory to be characterized morphologically, biochemical tests, and physiological reactions. Furthermore, their enzyme potential and resistance to heavy metals Pb, Cd, and Fe were tested. Dendogram construction using MVSP(Multi-Variate Statistical Package) software with isolated grouping based on the Unweighted Pair Group Method Averages (UPGMA) algorithm. The similarity between isolates using the Simple Matching Coefficient (SSM) similarity value. The results of dendogram construction obtained 3 clusters namely A (4 isolates), B (1 isolate), and C (2 isolates). The similarity index range between isolates is 0,543 – 0,857. Clusters A, B, and C have similarity indices ≤0.70 indicating that each cluster member is a distinct species.
