Abstract

Daun dewa (Gynura pseudochina) merupakan tanaman herbal yang memiliki berbagai khasiat karena banyak mengandung metabolit sekunder. Namun demikian, pola budidaya tanaman daun dewa secara konvensional menyebabkan ketidakseragaman kuantitas metabolit yang dihasilkan. Oleh karena itu, teknologi kultur jaringan dapat digunakan sebagai alternatif perbanyakan tanaman yang mampu menjamin keseragaman genetik. Salah satu teknologi kultur jaringan yang dapat digunakan adalah induksi kalus. Induksi kalus merupakan tahap awal dari teknik kultur in vitro yang bertujuan untuk menghasilkan dan memperbanyak sel kalus secara massal. Kalus adalah kumpulan sel yang aktif membelah karena adanya perlukaan. Perkembangan kalus dipengaruhi oleh zat pengatur tumbuh (ZPT) yang ditambahkan ke dalam media kultur. Penelitian mengenai pengaruh ZPT pada kultur induksi kalus tanaman G. pseudochina masih terbatas. Penelitian ini bertujuan untuk mengetahui pengaruh kombinasi konsentrasi 2,4-D (2,4-Dicholorophenoxyacetic acid) dan BAP (6-Benzyl Amino Purine) dalam menginduksi kalus daun dewa (G. pseudochina) secara in vitro. Penelitian ini menggunakan rancangan acak lengkap (RAL) faktorial yang terdiri dari dua faktor yaitu 2,4-D dan BAP. Masing-masing faktor tersebut terdiri dari empat taraf konsentrasi yaitu 0; 0,5; 1; dan 1,5 mg/L. Data dianalisis menggunakan Analysis of Variance (ANOVA) two-way dan dilanjutkan dengan post hock tukey jika terdapat hasil yang signifikan. Beberapa parameter yang diamati diantaranya adalah hari munculnya kalus, persentase kalogenesis, berat basah kalus, dan morfologi kalus. Hasil penelitian menunjukkan bahwa pemberian kombinasi konsentrasi 2,4-D dan BAP yang optimum dalam menginduksi kalus secara in vitro yaitu perlakuan 2,4-D 0,5 mg/L + BAP 0,5 mg/L dengan ditandai waktu muncul kalus paling cepat 12 hari, persentase kalogenesis 100%, rerata berat basah 1.12 g, warna kalus hijau, dan bertekstur remah.

Dewa leaf (Gynura pseudochina) is herbal plant that has various properties because it contains many secondary metabolites. However, the conventional cultivation pattern of dewa leaves causes the quantity of metabolites produced to be non-uniform. Therefore, tissue culture technology can be used as an alternative to plant propagation that can guarantee genetic uniformity. One of the tissue culture technologies that can be used is callus induction. Callus induction is the initial stage of in vitro culture technique which aims to produce and multiply callus cells en masse. Callus is a collection of cells that are actively dividing due to injury. Callus development is influenced by growth regulators added to the culture medium. Research on the effect of ZPT on callus-induced culture of G. pseudochina is still limited. This study aims to determine the effect of a combination of concentrations of 2,4-D (2,4-Dicholorophenoxyacetic acid) and BAP (6-Benzyl Amino Purine) to induce the formation of callus leaves of the gods (G. pseudochina) in vitro. This study used a factorial completely randomized design consisting of two factors, 2,4-D and BAP. Each of these factors consists of four concentration levels, 0; 0.5; 1; and 1.5 mg/L. Data were analyzed using a two-way Analysis of Variance (ANOVA) and followed by a post hock tukey if there were significant results. Some of the parameters observed included the day of callus emergence, the percentage of callogenesis, callus wet weight, and callus morphology. The results showed that giving the optimum combination of 2,4-D and BAP concentrations in inducing callus in vitro was the 2,4-D 0.5 mg/L + BAP 0.5 mg/L treatment with the fastest callus appearing time 12 days, the percentage of callogenesis was 100%, the average fresh weight was 1.12 g, the color of the callus was green, and the texture was friable.
