Abstract

Bakteri kitinolitik merupakan bakteri yang dapat menghidrolisis kitin menjadi turunannya secara enzimatis dengan enzim kitinase. Bakteri kitinolitik dapat ditemukan di limbah cangkang kepiting atau limbah udang. Karakter genus atau spesies bakteri dapat diketahui melalui pengamatan morfologi, uji biokimia, dan reaksi fisiologis. Berdasarkan karakter bakteri tersebut dapat dipelajari keanekaragamannya, salah satunya dengan metode pendekatan taksonomi numerik fenetik. Tujuan penelitian adalah mengetahui keanekaragaman bakteri kitinolitik yang diisolasi dari limbah cangkang kepiting dengan metode pendekatan taksonomi numerik fenetik. Isolasi dan skrining bakteri kitinolitik menggunakan media kitin didapatkan 5 isolat yaitu CK.7, CK.12, CK.13, CK.15, dan CK.22. Karakter yang didapat ditabulasikan ke dalam tabel n x t dan konstruksi dendogram menggunakan aplikasi Microsoft Excel dan program MVSP (Multivariate Statistical Package) versi 3.22. Analisis kekerabatan korelasi kofenetik dianggap realistis dan dapat diterima apabila nilai R > 0,7. Indeks kitinolitik dari kelima isolat tersebut antara 1,23 hingga 5,10. Dendogram yang dihasilkan dapat diterima dan realistis dengan nilai R sebesar 0,92. Dari dendogram tersebut terbentuk dua cluster yakni Cluster A (CK.13 dan CK.15) dan Cluster B (CK.7, CK.12, dan CK.22). Berdasarkan Generic Assignment dengan metode profile matching menggunakan panduan dari Bergey’s manual of determinative bacteriology didapatkan bahwa genus yang teridentifikasi adalah genus Veillonella (CK.13 dan CK.15) dan genus Vibrio (CK.7, CK. 12, dan CK.22).

Chitinolytic bacteria are bacteria that can hydrolyse chitin into its derivatives enzymatically with the enzyme chitinase. Chitinolytic bacteria can be found in crab shell waste or shrimp waste. The character of the genus or species of bacteria can be known through morphological observations, biochemical tests, and physiological reactions. Based on the character of the bacteria, its diversity can be studied, one of which is the method of phenetic numerical taxonomy approach. The purpose of the study was to determine the diversity of chitinolytic bacteria isolated from crab shell waste using the phenetic numerical taxonomy approach method. Isolation and screening of chitinolytic bacteria using chitin media obtained 5 isolates namely CK.7, CK.12, CK.13, CK.15, and CK.22. The characters obtained were tabulated into the n x t table and dendrogram construction using Microsoft Excel application and MVSP (Multivariate Statistical Package) version 3.22. Co-phenetic correlation kinship analysis is considered realistic and acceptable if the R value is > 0.7. The chitinolytic index of the five isolates ranged from 1.23 to 5.10. The resulting dendrogram is acceptable and realistic with an R value of 0.92. From the dendrogram, two clusters were formed, namely Cluster A (CK.13 and CK.15) and Cluster B (CK.7, CK.12, and CK.22). Based on Generic Assignment with profile matching method using guidelines from Bergey's manual of determinative bacteriology, it was found that the genus identified was the genus Veillonella (CK.13 and CK.15) and the genus Vibrio (CK.7, CK. 12, and CK.22).
