Abstract
Yeast adalah mikroorganisme eukariotik uniseluler yang memiliki metabolisme kemoorganotrof. Metabolisme yeast menghasilkan adenosin trifosfat (ATP) yang berguna untuk energi gerak, pertumbuhan dan biosintetis. Salah satu jalur metabolisme yeast adalah fermentasi. Fermentasi merupakan proses yang memecah molekul organik besar menjadi molekul sederhana yang terdiri alkohol dan asam organik. Saccharomyces cerevisiae (BTCC003) dan Hyphopichia burtonii (InaCCY066) dapat melakukan fermentasi glukosa dan xilosa. Spesies H. burtonii masih jarang diteliti sebagai agen fermentasi xilosa menjadi etanol dan xilitol. Tujuan penelitian adalah untuk mengetahui pertumbuhan sel, konsentrasi gula reduksi, konsentrasi gula total, dan penggunaan substrat (Yx/s) dari isolat BTCC003 dan isolat InaCCY066 dalam media Yeast extract-Peptone-Glucose (YPG), Yeast extract-Peptone-Xylose (YPX) dan Yeast extract-Peptone-Glucose-Xylose (YPGX). Fermentasi dilakukan dalam 75 ml media yang mengandung sel dengan OD 0,3 sebagai agen fermentasi dan diinkubasikan pada suhu 30ᵒC dengan agitasi 150 rpm dengan 3 ulangan. Isolat yang digunakan secara tunggal dan campuran dimana kedua isolat tersebut tidak saling antagonis berdasarkan zona hambat pertumbuhan. Pertumbuhan sel yang terdiri dari (1) fase adaptasi; (2) fase eksponensial; (3) fase stasioner; dan (4) fase kematian diukur dengan spektrofotometer pada OD600nm pada jam ke-0,3,6,24,48, dan 72, kandungan gula reduksi dengan metode DNS dan kadar gula total dengan metode Phenol-Sulfuric Acid. Hasil penelitian menunjukkan pertumbuhan isolat uji berbeda pada setiap jenis media yang digunakan. Isolat BTCC003 memiliki fase 1 dan 2 pada media YPG; fase 1, 2 dan 4 pada media YPX; fase 1,2 dan 3 pada media YPGX. Isolat InaCCY066 memiliki fase 1 dan 2 pada media YPG; fase 1, 2 dan 3 pada media YPX; fase 1,2 dan 4 pada media YPGX. Isolat campuran (BTCC003 + InaCCY066) memiliki fase 2 pada media YPG; fase 1,2,3, dan 4 pada media YPX; fase 2 pada media YPGX. Pada ketiga isolat (BTCC003, InaCCY066 dan campuran) mengalami penurunan konsentrasi gula reduksi pada media YPG dan habis tak bersisa sampai akhir pengamatan. Sedangkan, konsentrasi gula reduksi YPX dan konsentrasi gula total YPGX mengalami penurunan dan masih tersisa sampai akhir pengamatan. Penggunaan substrat (Yx/s) isolat BTCC003 pada media YPG sebanyak 0,75 (g/g), media YPX sebanyak 0,25 (g/g) dan media YPGX sebanyak 0,47 (g/g). Penggunaan substrat (Yx/s) isolat InaCCY066 pada media YPG sebanyak 0,45 (g/g), media YPX sebanyak 0,24 (g/g) dan media YPGX sebanyak 0,27 (g/g). Penggunaan substrat (Yx/s) isolat campuran pada media YPG sebanyak 1,70 (g/g), media YPX sebanyak 3,46 (g/g) dan media YPGX sebanyak 0,89 (g/g).

Yeast is a unicellular eukaryotic microorganism that has a chemoorganotrophic metabolism. Yeast metabolism produces Adenosine Triphosphate (ATP) which is useful for yeast motion energy, growth, and biosynthesis. One of the pathways of yeast metabolism is fermentation. Fermentation is a process that breaks down large organic molecules into simple molecules containing alcohol and organic acid. Saccharomyces cerevisiae (BTCC003) and Hyphopichia burtonii (InaCCY066) can ferment glucose and xylose. The species H. burtonii species is rarely studied as xylose fermentation agent. The purpose of this research is to determine the growth and maximum specific growth rate (µmax) of isolate BTCC003 and isolate InaCCY066 in the media of Yeast extract-Peptone-Glucose (YPG), Yeast extract-Peptone-Xylose (YPX) dan Yeast extract-Peptone-Glucose-Xylose (YPGX). Fermentation was carried out in 75 ml of media containing cells with OD 0,3 as the agent of fermentation and incubated at 30ᵒC with agitation of 150 rpm with 3 replications. The isolats used singly and mixtures where two isolats have been tested for antagonist by measuring the zone of growth inhibition. Cell growth consisting of (1) lag phase; (2) log phase; (3) stationary phase; and (4) the death phase was measured by a spectrophotometer at OD600nm at 0,3,6,24,48, and 72 hours, reducing sugar content by DNS method and total sugar content by Phenol-Sulfuric Acid method. The result of this research is growth of the isolate of the test differs from any type of media used. Isolate BTCC003 has phases 1 and 2 on YPG media; phases 1, 2 and 4 on YPX media; phases 1, 2 and 3 on YPGX media. Isolate InaCCY066 has phases 1 and 2 on YPG media; phases 1, 2 and 3 on YPX media; phases 1, 2 and 4 on YPGX media. The mixed isolate (BTCC003 + InaCCY066) had phase 2 on YPG media; phases 1,2,3, and 4 on YPX media; phase 2 on YPGX media. The three isolates (BTCC003, InaCCY066 and mixture), experienced a decrease in the concentration of reducing sugars in YPG media and were completely used up until the end of the observation. The concentration reducing sugar in YPX and total sugar concentration YPGX decreased and remained until the end of the observation. The use of substrate (Yx/s) isolate BTCC003 on YPG media was 0,75 (g/g), YPX media was 0,25 (g/) and YPGX media was 0,47 (g/g). The use of substrate (Yx/s) isolate InaCCY066 on YPG media was 0,45 (g/g), YPX media was 0,24 (g/) and YPGX media was 0,27 (g/g). The use of substrates (Yx/s) of mixed isolates on YPG media was 1,70 (g/g), YPX media was 3,46 (g/g) and YPGX media was 0,89 (g/g).
