Abstract

Deteksi air mani dalam ilmu forensik sangat penting dalam kasus kekerasan seksual dugaan persetubuhan. Kristal kolin dapat diketahui dalam konsentrasi tinggi pada cairan mani menggunakan pengujian Florance iodine (FI). Uji FI ini telah digunakan dalam ilmu forensik selama bertahun-tahun, namun sangat sedikit yang didokumentasikan mengenai optimalisasi, ketahanan dan masa kadaluwarsa reagen FI tersebut dalam pekerjaan kasus forensik. Penelitian ini bertujuan untuk mengetahui optimalisasi konsentrasi iodin pada Reagen FI termasuk dengan ketahanan dan masa kadaluwarsa reagen FI tersebut. Uji optimalisasi konsentrasi didapatkan bahwa konsentrasi optimum iodine pada reagen FI adalah 10%, didapatkan pula ketahanan reagen FI pada pH asam adalah 1 dan pada pH basa adalah 8 dan didapatkan masa kadaluarsa reagen FI adalah selama 7 minggu, dan dilakukan pengujian FT IR dan Raman pada Reagen FI minggu ke-0 dan minggu ke-7 dan didapatkan hasil bahwa terdapat perbedaaan yang signifikan pada intensitas kadar Iodine yang terdapat pada reagen FI.

Detection of semen in forensic science is very important in cases of alleged sexual assault. Choline crystals can be detected in high concentrations in seminal fluid using the Florance Iodine (FI) test for detection. The Florence test has been used in forensic science for many years, but very little has been documented regarding the optimization, durability and shelf-life of these FI reagents in forensic case work. This thesis aims to determine the optimization of the concentration of iodine in the FI reagent including the resistance and expiration of the FI reagent. The concentration optimization test found that the optimum concentration of iodine in the FI reagent was 10%, the resistance of the FI reagent at acidic pH was 1 and at alkaline pH was 8 and the expiration period of the FI reagent was 7 weeks, and FT- IR and Raman tests were performed. on the FI reagent week 0 and week 7 and it was found that there was a significant difference in the intensity of the Iodine levels contained in the FI reagent.
