Abstract

Antioksidan merupakan senyawa yang dapat melindungi sel-sel tubuh dari kerusakan akibat radikal bebas. Reaksi rantai radikal bebas dihentikan dengan memberikan elektron ke molekul radikal bebas dan mengubahnya menjadi molekul stabil yang tidak berbahaya bagi sel tubuh. Delima (Punica granatum) merupakan salah satu buah yang dapat dimanfaatkan sebagai nutraseutikal karena memiliki potensi aktivitas antioksidan yang tinggi. Pada penelitian ini digunakan kulit dan mesocarp yang merupakan bagian yang tidak dikonsumsi dari tiga sub-spesies yang memiliki warna kulit berbeda yaitu merah, putih dan ungu. Analisis yang akan dilakukan bertujuan untuk mendapatkan informasi mengenai tingkat antioksidan masing-masing bagian pada tiga sub-spesies delima. Kapasitas antioksidan didasarkan pada pengukuran TPC, TFC, ABTS (2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid) dan DPPH (2,2-diphenyl-1-picrylhydrazyl) melalui metode kolorimetri menggunakan spektrofotometer. Berdasarkan hasil uji didapatkan nilai kandungan total fenolik kulit delima ungu 721 mgGAE/g, kulit merah 192 mgGAE/g, dan putih 424 mgGAE/g. Total fenolik pada mesocarp ungu yaitu 338 mgGAE/g, merah 127 mgGAE/g, dan putih 357 mgGAE/g. Kandungan total flavonoid kulit ungu 161 mgQE/g, merah 34 mgQE/g, dan putih 82 mgQE/g. Kandungan total flavonoid mesocarp buah delima ungu 91 mgQE/g, merah 26 mgQE/g, dan putih 94 mgQE/g. Kulit memiliki aktivitas antioksidan lebih tinggi dari pada mesocarp karena lebih banyak ditemukan senyawa bioaktif pada kulit. Kulit dan mesocarp delima ungu memiliki aktivitas antioksidan sangat kuat diantara ketiga sub-spesies dengan nilai IC50 <50 μg/ml. Penelitian ini dapat digunakan sebagai informasi awal mengenai sumber baru antioksidan.

Antioxidants are compounds that can protect the body's cells from damage caused by free radicals. The free radical chain reaction is stopped by giving electrons to free radical molecules and turning them into stable molecules that are not harmful to the body's cells. Pomegranate (Punica Granatum L.) is one of the fruits that can be used as nutraseutical because it has a high potential antioxidant activity. In this study the peel and mesocarp are used which are parts that are not consumed from three sub-species that have different peel colors, namely red, white and purple. Analysis to be carried out aims to obtain information about the level of antioxidants of each part in three pomegranate sub-species. The antioxidant capacity is based on the measurement of TPC, TFC, ABTS (2,2'-Azino-BIS 3-Ethylbenzothiazoline-6-sulfonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl) through the colorimetric method using the spectrophotometer. Based on the test results obtained the value of the total phenolic content of purple pomegranate peel 721 mgGAE/g, red peel 192 mgGAE/g, and white 424 mgGAE/g. The total phenolic in the purple mesocarp is 338 mgGAE/g, red 127 mgGAE/g, and white 357 mgGAE/g. The total content of purple peel flavonoids 161mgQE/g, red 34 mgQE/g, and white 82 mgQE/g. The total content of the purple pomegranate mesocarp flavonoids 91 mgQE/g, red 26 mgQE/g, and white 94 mgQE/g. The peel has higher antioxidant activity than mesocarp because more bioactive compounds are found in the peel. Purple pomegranate peel and mesocarp have very strong antioxidant activity between the three sub-species with IC50 value <50 μg/ml. This research can be used as initial information about new sources of antioxidants.
