Abstract

Satu penyebab hiperpigmentasi adalah radiasi ultaviolet (UV) yang menyebabkan produksi ROS (Reactive Oxygen Species) sehingga memicu pembentukan melanin. Kondisi ini dapat diatasi dengan inhibitor enzim tirosinase, salah satunya dari senyawa flavonoid yang terkandung dalam buah parijoto (Medinilla speciosa). Tujuan penelitian ini adalah mengukur aktivitas antioksidan dan penghambatan tirosinase ekstrak parijoto serta mengetahui efek pemberian ekstrak parijoto 25% terhadap kulit mencit (Mus musculus) yang diinduksi UV. Metode uji in-vitro menggunakan analisis DPPH untuk antioksidan dan dopakrom untuk penghambatan tirosinase. Sedangkan metode in-vivo dilakukan dengan hewan uji untuk mengukur kadar ROS, melanin, dan ekspresi protein yang berkaitan dengan melanogenesis. Hasil yang diperoleh menunjukkan ekstrak parijoto merupakan inhibitor sangat lemah dalam menghambat protein tirosinase dengan nilai IC50 tidak teridentifikasi meskipun aktivitas antioksidan pada ekstrak parijoto bernilai IC50 4,924 μg/mL atau tergolong sangat kuat. Antioksidan berperan langsung dalam menghambat terjadinya melanogenesis. Adapun hasil uji kadar ROS, melanin, dan ekspresi protein tirosinase pada kulit mencit menunjukkan nilai masing-masing 28,83 U/L; 53 sel; dan 8,761 pixel, berbeda secara signifikan (p < 0,05) dengan kontrol negatif dan positif yang bernilai masing-masing 36,30 U/L, 141 sel, dan 53,041 pixel serta 41,17 U/L; 148 sel; dan 52,870 pixel. Ini menunjukkan bahwa ekstrak parijoto secara in-vivo mampu bertindak sebagai antihiperpigmentasi.

One cause of hyperpigmentation is ultraviolet (UV) radiation which causes the production of ROS (Reactive Oxygen Species) which triggers the formation of melanin. This condition can be overcome with anti-hyperpigmentation agents that inhibit the performance of the tyrosinase enzyme, one of which is the flavonoid compound contained in Parijoto fruit (Medinilla speciosa). The purpose of this study was to measure the antioxidant activity and tyrosinase inhibition of parijoto extract and to determine the effect of giving 25% parijoto extract on UV-induced skin of mice (Mus musculus). The in-vitro test method uses DPPH analysis for antioxidants and dopachrome for tyrosinase inhibition. Meanwhile, the in-vivo method is carried out with test animals to measure levels of ROS, melanin, and protein expression related to melanogenesis. The results obtained show that parijoto extract is a very weak inhibitor in inhibiting protein tyrosinase with an IC50 value not identified even though the antioxidant activity of parijoto extract has an IC50 value of 4.924 μg/mL or is classified as very strong. Antioxidants play a direct role in inhibiting melanogenesis. The test results for ROS levels, melanin, and tyrosinase protein expression in mouse skin showed values of 28.83 U/L respectively; 53 cells; and 8,761 pixels, significantly different (p < 0.05) from the negative and positive controls which were respectively 36.30 U/L, 141 cells, and 53,041 pixels and 41.17 U/L; 148 cells; and 52,870 pixels. This shows that in-vivo parijoto extract is able to be an antihyperpigmentation agent.
