Abstract

Paparan sinar UV merupakan salah satu pemicu hiperpigmentasi, keadaan dimana terjadi produksi melanin secara berlebihan sehingga menyebabkan spot, rona maupun titik kehitaman pada jaringan kulit. Untuk mengatasi hiperpigmentasi dapat dilakukan terapi topikal maupun oral menggunakan agen antioksidan untuk menurunkan aktivitas tirosinase maupun dengan terapi laser. Salah satu agen anti hiperpigmentasi potensial adalah trisindolina 1 yang mampu menghambat aktivitas tirosinase. Pada penelitian ini dilakukan uji ekspresi melalui metode qRT-PCR untuk mengetahui ekspresi dari gen MC1R dan MITF yang bersifat pro hiperpigmentasi. Pada penelitian ini juga dilakukan pewarnaan hematoksilin eosin pada jaringan dorsal Mus musculus untuk mengetahui sebaran melanosit pada mencit yang dipapar oleh sinar UV-B. Hasil dari penelitian diperoleh peningkatan ekspresi dari gen MC1R maupun MITF masing 2.43x pada gen MITF dan 6.7x pada gen MC1R. Sebaran melanosit pada mencit dengan induksi trisindolina 1 berada pada kategori ‘+’ (sedikit) apabila dibandingkan dengan kontrol ‘+++’ (tinggi). Kesimpulan dari penelitian ini adalah Trisindolina 1 mampu mengurangi sebaran melanosit pada jaringan kulit namun masih belum efektif untuk mereduksi ekspresi dari gen pro hiperpigmentasi.

UV rays exposure is one of cause for hyperpigmentation, a condition of excessive melanin production, causing black spots, tints or dots on the skin tissue. Hyperpigmentation usually treated using antioxidant agent through topical or oral therapy to reduce tyrosinase activity or laser therapy. Promising novel compound with anti-hyperpigmentation properties is trisindoline 1 which can inhibit tyrosinase activity. In this study, gene expression assay was conducted using qRT-PCR to determine the expression of MC1R and MITF that acts as pro hyperpigmentation gene. Hematoxylin-eosin staining also conducted to analyse the melanocyte distribution in mice exposed to UV B. From the research that has been carried out, it was found that the expression of the pro hyperpigmentation genes is increased. The expression of MITF gene is increased by 2.43 fold moreover the expression of MC1R gene are also increased by 6.7 fold. The distribution of melanocytes in mice with trisindoline 1 induction was in the '+' (few) category when compared with '+++' (high) in the control group. The conclusion of this research is that Trisindolina 1 can reduce the distribution of melanocytes in skin tissue but was not effective enough in reducing the expression of pro-hyperpigmentation genes.
