Abstract

Kalium sangat diperlukan oleh tanaman untuk merangsang pertumbuhan dan perkembangan buah. Efek pemberian kalium eksogen dalam meningkatkan kualitas buah, dapat dilihat dari kandungan fitokimianya seperti kadar pigmen klorofil dan karotenoid. Tujuan dari penelitian ini adalah mengetahui pengaruh aplikasi kalium eksogen secara soluble dan foliar terhadap kadar klorofil, karoten α- dan β, serta ekspresi gen CmChlG dan CmPSY pada buah melon kultivar GMP (Gama Melon Parfum). Pengukuran kadar klorofil dan karoten digunakan metode spektrofotometer sedangkan ekspresi gen digunakan metode Reverse Transcript-quantitative Polymerase Chain Reaction (RT-qPCR). Analisis data kadar klorofil, karoten α- dan β- dilakukan dengan uji ANOVA One Way (p<95%) sedangkan analisis gen dilakukan secara deskriptif. Hasil pengamatan menunjukkan pemberian kalium eksogen berpengaruh nyata terhadap kandungan total klorofil dan karoten α- dan β pada buah melon kultivar GMP. Kalium eksogen secara soluble mengakibatkan penurunan kadar total klorofil sebanyak 0,1087 gr/mL FW, α-karoten sebanyak 0,066 gr/mL FW dan β-karoten sebanyak 0,0781 gr/mL FW. Sementara itu kalium eksogen secara foliar mengakibatkan peningkatan kadar total klorofil sebanyak 0,2234 gr/mL FW, α-karoten sebanyak 0.0809 gr/mL FW dan β- karoten sebanyak 0,0809 gr/mL FW. Eskpresi gen CmChlG dan CmPSY juga mengalami perubahan pada kedua perlakuan penambahan kalium. Ekspresi gen CmChlG pada perlakuan foliar, mengalami peningkatan sekitar 0,407 fold change kemudian untuk perlakuan soluble mengalami penurunan 0,179 fold change. Sementara itu, ekspresi gen CmPSY pada perlakuan foliar, mengalami peningkatan sekitar 1,805 fold change sedangkan perlakuan soluble menunjukkan penurunan sekitar 0,437 fold change. Hasil ekspresi gen tersebut berkorelasi positif dengan hasil pengujian kadar pigmen klorofil dan karotenoid.

Potassium is needed by plants to stimulate the growth and development of fruit. The effect of exogenous potassium in improving fruit quality can be seen from the phytochemical content such as levels of chlorophyll and carotenoid pigments. The aim of this research was to determine the effect of soluble and foliar application of exogenous potassium on the levels of chlorophyll, α- and β-carotene, as well as the expression of the CmChlG and CmPSY genes in the melon cultivar GMP (Gama Melon Parfum). Measuring chlorophyll and carotene levels used the spectrophotometer method, while gene expression used the Reverse Transcriptquantitative Polymerase Chain Reaction (RT-qPCR) method. Data analysis of chlorophyll, α-and β-carotene levels was carried out using the One Way ANOVA test (p<95%) while gene analysis was carried out descriptively. The observation results showed that the administration of exogenous potassium had a significant effect on the total content of chlorophyll and α- and β-carotene in the melon cultivar GMP. Soluble exogenous potassium resulted in a decrease in total chlorophyll levels by 0.1087 gr/mL FW, α-carotene by 0.066 gr/mL FW and β-carotene by 0.0781 gr/mL FW. Meanwhile, foliar exogenous potassium resulted in an increase in total chlorophyll levels by 0.2234 gr/mL FW, α-carotene by 0.0809 gr/mL FW and β-carotene by 0.0809 gr/mL FW. The expression of the CmChlG and CmPSY genes also changed in the two potassium addition treatments. The expression of the CmChlG gene in the foliar treatment increased by around 0.407 fold change, then for the soluble treatment it decreased by 0.179 fold change. Meanwhile, the expression of the CmPSY gene in the foliar treatment increased by around 1.805 fold change, while the soluble treatment showed a decrease of around 0.437 fold change. The results of gene expression were positively correlated with the results of testing levels of chlorophyll and carotenoid pigments.
