Abstract
Metilen biru (MB) adalah salah satu zat warna yang umum digunakan dalam industri tekstil karena relatif murah dan mudah diperoleh. Akan tetapi, pembuangan limbah cair MB dapat menimbulkan pencemaran lingkungan dan mengganggu kehidupan ekosistem. Untuk mengatasi hal tersebut, biodegradasi menggunakan mikroorganisme merupakan salah satu metode yang dapat dilakukan. Tujuan dari penelitian ini adalah untuk menganalisis kemampuan degradasi pewarna MB oleh Pseudomonas aeruginosa yang terimobilisasi pada matriks SA/PVA/UiO-66. Hasil penelitian menunjukkan beads SA/PVA/UiO-66 terimobilisasi P. aeruginosa yang dipreparasi menggunakan crosslinker komposit CaCl2/asam borat memiliki persentase dekolorisasi tertinggi sebesar 92,28%. Hasil studi kinetika dan isotermal adsorpsi menyebutkan adsorpsi mengikuti pola orde dua semu dan isoterm Langmuir. Melalui optimasi dengan metode RSM, diperoleh kondisi optimal proses biodekolorisasi MB pada pH 7,40; suhu 36,16 °C; dan waktu inkubasi 54,82 jam dengan hasil persentase dekolorisasi MB 96,54%. Senyawa metabolit yang dihasilkan pada proses biodekolorisasi MB diprediksi terdiri atas azure A, azure B, leuko-MB, dan 3-((3-(dimetilamino) fenil) sulfinil)-N1,N1-dimetilbenzen-1,4-diamina. Selain itu, uji aktivitas lakase dan mangan peroksidase yang merupakan enzim ligninolitik dari P. aeruginosa masing-masing menunjukkan aktivitas sebesar 2,60 x 102 dan 1,65 x 103 U/mL

Methylene blue (MB) is one of the dyes that is commonly used in the textile industry because it is relatively cheap and easy to obtain. Nevertheless, the disposal of MB wastewater can cause environmental pollution and disrupt the ecosystem life. To overcome this, biodegradation using microorganisms is one of the methods that can be carried out. The aim of this study was to analyze the degradation ability of MB by bacterium Pseudomonas aeruginosa immobilized on the SA/PVA/UiO-66 matrix. The results showed that P. aeruginosa immobilized SA/PVA/UiO-66 beads prepared using composite of CaCl2/boric acid crosslinkers had the highest decolorization percentage (92.28%.). Moreover, the kinetics and adsorption isotherm studies showed that adsorption follows a pseudo-second-order model and the Langmuir isotherm. Through optimation using the RSM method, optimal conditions for the MB biodecolorization process were obtained at pH 7.40, temperature 36.16 °C, and incubation time of 54.82 hours with a resulting MB decolorization percentage of 96.54%. The metabolites produced in the MB biodecolorization process were predicted to consist of azure A, azure B, leuco-MB,
and 3-((3-(dimethylamino) phenyl) sulfinyl)-N1,N1-dimethylbenzen-1,4-diamine. Apart from that, the results of laccase and manganese peroxidase activity tests (ligninolytic enzymes produced by P. aeruginosa) were 2.65 x 102 dan 1.65 x 103 U/mL, respectively.
