Abstract

isang Kayu (M. paradiasica L. Var. Kayu) digunakan oleh masyarakat Desa Senduro, Lumajang, Jawa Timur, sebagai antidiare. Standardisasi farmasitikal diperlukan untuk mengembangkan obat tradisional menjadi obat herbal terstandar dan fitofarmaka, melalui tahapan pengujian keamanan, khasiat preklinik (in vitro dan in vivo), dan klinik. Salah satu uji preklinik adalah pengujian hematologi, meliputi jumlah leukosit total, jumlah leukosit diferensial, konsentrasi hemoglobin, dan jumlah eritrosit total. Penelitian ini bertujuan mengetahui pengaruh ekstrak etanol buah mentah pisang kayu pada hematologi mencit betina (M. musculus). Ekstraksi dilakukan dengan maserasi untuk mengeluarkan senyawa kimia yang terkandung dalam buah mentah pisang kayu. Perhitungan leukosit dan eritrosit total menggunakan counting chamber hemositometer, kadar hemoglobin dengan Hb Meter, dan uji diferensial leukosit dengan pewarnaan Giemsa 1% pada apusan darah. Mencit betina dibagi menjadi 7 kelompok: kontrol aquades 0,5 ml, kontrol CMC-Na 0,2 ml, dan kelompok perlakuan dengan dosis ekstrak etanol buah pisang kayu 50 mg/kgBB, 100 mg/kgBB, 200 mg/kgBB, 400 mg/kgBB, dan 800 mg/kgBB. Analisis statistik dilakukan dengan One-Way ANOVA. Hasil penelitian menunjukkan pemberian ekstrak etanol buah mentah pisang kayu tidak berpengaruh nyata (p>0,05) pada jumlah eritrosit total, kadar hemoglobin, jumlah leukosit total, dan leukosit diferensial (neutrofil, limfosit, monosit, eosinofil, dan basofil) antar kelompok perlakuan. Seluruh rata-rata pada komponen darah yang diuji berada pada kisaran parameter normal, sehingga dapat disimpulkan bahwa ekstrak etanol buah mentah pisang kayu (M. paradisiaca L. var. Kayu) tidak mempengaruhi hematologi hewan uji M. musculus.

Banana Kayu (Musa paradisiaca L. Var. Kayu) is used by the community in Senduro Village, Lumajang, East Java, as an anti-diarrheal remedy. Pharmaceutical standardization is required to develop traditional medicine into standardized herbal medicine and phytopharmaceuticals, through stages of safety testing, preclinical efficacy (in vitro and in vivo), and clinical testing. One preclinical test is hematology testing, including total leukocyte count, differential leukocyte count, hemoglobin concentration, and total erythrocyte count. This study aims to determine the effect of etanol extract of unripe Banana Kayu on the hematology of female mice (M. musculus). Extraction is carried out by maceration to release the chemical compounds contained in the raw fruit of the unripe Banana Kayu. Total leukocyte and erythrocyte counts were performed using a counting chamber hemocytometer, hemoglobin levels with an Hb Meter, and differential leukocyte test with 1% Giemsa staining on blood smears. Female mice were divided into 7 groups: control with 0.5 ml aquades, control with 0.2 ml CMC-Na, and treatment groups with etanol extract of Pisang Kayu fruit at doses of 50 mg/kgBW, 100 mg/kgBW, 200 mg/kgBW, 400 mg/kgBW, and 800 mg/kgBW. Statistical analysis was performed using One-Way ANOVA. The results showed that administration of etanol extract of unripe Pisang Kayu fruit had no significant effect (p>0,05) on total erythrocyte count, hemoglobin levels, total leukocyte count, and differential leukocytes (neutrophils, lymphocytes, monocytes, eosinophils, and basophils) between treatment groups. All average data on the tested blood components were within the normal parameter range, concluding that the etanol extract of unripe Banana Kayu fruit (Musa paradisiaca L. Var. Kayu) does not affect the hematology of M. musculus test animals.
