Abstract

Demam berdarah dengue (DBD) merupakan masalah kesehatan global yang semakin mendesak, dengan risiko infeksi yang terus meningkat. Deteksi dini dan spesifik virus dengue angat penting untuk menghindari kesalahan diagnosis dan penundaan pengobatan yang dapat meningkatkan angka kematian. Metode NS1 dianggap efektif dan efisien dalam mendeteksi virus dengue pada periode awal infeksi, namun membutuhkan selektivitas tinggi untuk kinerja optimal. Penelitian ini mengevaluasi sensitivitas dan selektivitas antara material CIS/ZnS nanopartikel yang terkonjugasi biotin dan antigen NS1 virus dengue, menggunakan quenching fluorescent untuk menentukan kinerja deteksi. Penelitian diawali dengan sintesis CIS/ZnS nanopartikel menggunakan metode wet chemical dan hot injection yang menghasilkan emisi kuning-oranye, dan biokonjugasi streptavidin dan antibodi NS1 biotin memungkinkan interaksi spesifik dengan antigen virus dengue. Sensitivitas material nanoprobe biomarker NS1 glikoprotein antigen virus dengue dapat dilihat melalui efisiensi quenching fluorescent yang tinggi, nilai LoD dan LoQ, dan linearitas quenching terhadap konsentrasi. Efisiensi quenching fluorescent material nanoprobe ini diuji pada konsentrasi antigen dengue 220 nM, 260 nM, dan 300 nM dalam media phosphate buffer solution (PBS) dan human plasma serum (HPS). Hasil menunjukkan bahwa efisiensi quenching fluorescent meningkat secara signifikan terhadap konsentrasi antigen dalam PBS dan HPS. Keberadaan protein tambahan dalam HPS mempengaruhi hasil deteksi dengan mengubah interaksi antara nanoprobe dan molekul target. Selektivitas atau interaksi spesifik material nanoprobe dengan antigen dinilai dari nilai efisiensi quenching fluorescent. Material CIS/ZnS-SA-Ab menunjukkan interaksi spesifik dengan antigen NS1, yang ditandai dengan kenaikan intensitas photoluminescence, serta nilai quenching fluorescent positif. Sebaliknya, tidak ada interaksi spesifik dengan antigen influenza dengan penurunan intensitas photoluminescence, mengindikasikan selektivitas tinggi dari nanoprobe terhadap antigen dengue.

Dengue fever (DF) is a serious global health issue, with infections predicted to increase. Early and specific detection of the dengue virus is crucial to avoid diagnostic errors and treatment delays that can raise mortality rates. The NS1 method has been considered effective and efficient for detecting the dengue virus during the early stages of infection, but it requires high selectivity for optimal performance. This study evaluates the sensitivity and selectivity of CIS/ZnS nanoparticle materials conjugated with biotin and dengue virus NS1 antigen, using fluorescent quenching to assess detection performance. The research began with the synthesis of CIS/ZnS nanoparticles using wet chemical and hot injection methods, which resulted in yellow-orange emission. Bioconjugation of streptavidin and NS1 biotin antibodies allowed specific interactions with the dengue virus antigen. The sensitivity of the NS1 biomarker nanoprobe material is evident from high fluorescent quenching efficiency, LoD and LoQ values, and quenching linearity to concentration. The fluorescent quenching efficiency of this nanoprobe material was tested at dengue antigen concentrations of 220 nM, 260 nM, and 300 nM in phosphate buffer solution (PBS) and human plasma serum (HPS). Results showed that the fluorescent quenching efficiency significantly increased with the antigen concentration in PBS and HPS. The presence of additional proteins in HPS affected the detection results by altering the interaction between the nanoprobe and target molecules. The selectivity or specific interaction of the nanoprobe material with the antigen is assessed by the efficiency of fluorescent quenching. CIS/ZnS-SA-Ab material demonstrates specific interaction with the NS1 antigen, indicated by an increase in photoluminescence intensity and positive fluorescent quenching values. In contrast, there is no specific interaction with influenza antigen, as evidenced by a decrease in photoluminescence intensity, indicating high selectivity of the nanoprobe for dengue antigens.
