Abstract

Salah satu penyakit yang paling banyak mempengaruhi produktivitas ikan lele (C. gariepinus) adalah Enteric Septicaemia of Catfish (ESC) yang disebabkan oleh bakteri E. ictaluri. Penyakit ini menyebabkan kerusakan di organ vital serta kematian ikan lele hingga 50%. Pengobatan dilakukan menggunakan berbagai jenis antibiotik yang memberikan efek samping residu antibiotik penyebab resistensi. Banyak dikembangkan alternatif pengobatan yang aman bagi kesehatan konsumen, salah satunya penggunaan probiotik. Penelitian sebelumya diketahui W. confusa memiliki potensi dalam menangani E. ictaluri secara in vitro. Oleh karena itu, pada penelitian ini dilakukan pengamatan respon imun C. gariepinus yang diinfeksi E. ictaluri setelah pemberian pakan tersuplementasi W. confusa selama 30 hari dengan beberapa dosis perlakuan (K1=10^4 CFU/mL, K2=10^6 CFU/mL dan K3=10^8 CFU/mL) untuk mempelajari pengaruhnya terhadap respon hematologi berupa kadar eritrosit, leukosit dan diferensial leukosit. Hasil penelitian menunjukkan bahwa suplementasi W. confusa berpengaruh nyata terhadap kadar eritrosit, leukosit, dan monosit pada C. gariepinus yang terinfeksi E. ictaluri. Secara khusus, pengaruh suplementasi bakteri probiotik W. confusa pada perlakuan K3 konsentrasi sebesar 10^8 CFU/mL yang memberikan pengaruh nyata (p<0,05) terhadap kontrol, pada jumlah rata-rata eritrosit eritrosit mendekati normal sebesar 1,294x10^6 sel/mm^3 dan kenaikan leukosit yang tidak jauh tinggi dari kadar leukosit normal yaitu, sebesar 309.090 sel/mm^3. Pada analisis diferensial leukosit didapatkan peningkatan monosit yang berpengaruh nyata terhadap kontrol pada perlakuan K2 dan K3 konsentrasi sebesar 10^6 CFU/mL dan 10^8 CFU/mL. Hasil penelitian ini menunjukkan suplementasi W.confusa pada pakan ikan dapat menjadi alternatif profilaksis dalam manajemen penyakit di lingkungan akuatik.

One of the most common diseases affecting catfish (C. gariepinus) productivity is Enteric Septicaemia of Catfish (ESC) caused by E. ictaluri bacteria. This disease causes damage to vital organs and catfish mortality of up to 50%. Treatment is carried out using various types of antibiotics that give side effects of antibiotic residues that cause resistance. Many alternative treatments have been developed that are safe for consumer health, one of which is the use of probiotics. Previous research found that W. confusa has the potential to treat E. ictaluri in vitro. Therefore, in this study, the immune response of C. gariepinus infected with E. ictaluri was observed after feeding W. confusa supplementation for 30 days with several treatment doses to study its effect on hematological responses in the form of erythrocyte, leukocyte and leukocyte differential levels. The results showed that W. confusa supplementation had a significant effect on the levels of erythrocytes, leukocytes, and monocytes in C. gariepinus infected with E. ictaluri. In particular, the effect of supplementation of probiotic bacteria W. confusa in the K3 treatment at a concentration of 10^8 CFU/mL which gave a significant effect (p<0.05) against the control, on the average number of erythrocytes close to normal at 1,294x10^6 cells/mm^3 and an increase in leukocytes which is not much higher than normal leukocyte levels, namely, 309.090 cells/mm^3. In the leukocyte differential analysis, it was found that the increase in monocytes had a significant effect on the control in the K2 and K3 treatments with concentrations of 10^6 CFU/mL and 10^8 CFU/mL. The results of this study indicate that W.confusa supplementation in fish feed can be an alternative prophylaxis in disease management in aquatic environments.
