Abstract
Temu kunci merupakan tanaman herbal yang termasuk dalam famili Zingiberaceae. Tanaman ini digunakan sebagai obat tradisional dan dimanfaatkan sebagai bahan rempah-rempah di negara Asia. Di Indonesia tanaman ini dimanfaatkan sebagai ”jamu” dan tonik tradisional yang digunakan dalam kecantikan. Potensi temu kunci sebagai obat tradisional berhubungan dengan senyawa metabolit sekunder yang terdapat didalamnya. Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder dari fraksi C2 ekstrak metanol rimpang temu kunci. Pada penelitian ini digunakan metode maserasi pada proses ekstraksi. Fraksinasi ekstrak temu kunci dilakukan dengan menggunakan meode Kromatografi Cair Vakum (KCV) dan Kromatografi Kolom Gravitasi (KKG), sedangkan uji pemurnian dilakukan dengan metode TLC 2D (2 dimensi) dan HPLC. Pada fraksinasi digunakan pelarut n-heksana, metilan klorida (MC), Etil Asetat (EA), dan Metanol (MeOH). Hasil isolasi yang telah diuji kemurniannya didapatkan senyawa C2 berbentuk padatan kristal dan berwarna kuning pucat. Identifikasi senyawa C2 yang berhasil diisolasi menggunakan Differential Scanning Calorimetry (DSC), Fourier Transform Infra-red (FTIR), dan Nuclear Magnetic Resonance (NMR) baik proton NMR dan 13C NMR. Senyawa C2 memiliki titik leleh 198,68°C. Data yang didapatkan dari analasis senyawa FTIR, proton NMR, dan 13C NMR dibandingkan dengan literatur yang telah dilaporkan sebelumnya. Hasilnya didapatkan bahwa senyawa C2 merupakan senyawa pinocembrin.

Temu Kunci is an herbal plant that belonging to the Zingiberaceae family. This plant is used as tradicional medicine and as spice in Asian countries. In Indonesia, temu kunci utilized in traditional herbal medicine called jamu and used as a beauty tonic. The potential of temu kunci as a traditional medicine is related to the secondary metabolite compounds it contains. This study was conducted to isolate secondary metabolite compounds from the fraction C2 methanol extract of temu kunci rhizomes. The milled dried rhizome of Temu Kunci was extracted with methanol using maceration method. Fractination of the Temu Kunci extract was performed using Vacum Liquid Chromatography (VLC) and Gravity Column Chromatography (GCC), while purification tests were conducted using 2-dimensional Thin Layer Chromatography (2D-TLC) and High-Performance Liquid Chromatography (HPLC). Solvents used in the fractionation process included n-hexane, methylene chloride (MC), ethyl acetate (EA), and methanol (MeOH). The isolation results, which were tested for purity, revealed that the C2 compound was obtained in the form of pale-yellow crystalline solids. The identification of the isolated C2 compound was carried out using Differential Scanning Calorimetry (DSC), Fourier Transform Infrared (FTIR) spectroscopy, and Nuclear Magnetic Resonance (NMR) spectroscopy, including both proton NMR and 13C NMR. The C2 compound had a melting point of 198.68°C. The data obtained from the FTIR, proton NMR, and 13C NMR analyses were compared with previously reported literature. The results indicated that the C2 compound is a Pinocembrin compound.
