Abstract

Tanaman Boesenbergia Rotunda disebut juga temu kunci adalah salah satu tanaman yang digunakan sebagai tanaman obat tradisional. Temu kunci berasal dari daerah Asia Tenggara yaitu dari negara Indonesia, Malaysia, dan Thailand. Tanaman ini digunakan secara tradisional untuk menyembuhkan penyakit tukak lambung dan obat batuk. Terdapat senyawa metabolit sekunder yang berpotensi sebagai agen antioksidan, antibakteri, anti inflamatori, dan anti mutagenik. Pada penelitian ini dilakukan isolasi senyawa metabolit sekunder dari rimpang temu kunci dari Ponorogo. Metode yang digunakan untuk mengisolasi senyawa yaitu ekstraksi maserasi dengan metanol menghasilkan 230,53 gram ekstrak kental dan pemisahan dengan kromatografi kolom. Hasil dari kromatografi cair vakum (KCV) yang dielusi menggunakan n-Heksana, diklorometana, etil asetat, dan metanol. Hasil pemisahan yaitu fraksi A, B, C, dan D. Fraksi C sebanyak 15 gram dipisahkan kembali menggunakan kolom kromatografi gravitasi (KKG) yang dielusi dengan n-Heksana : etil asetat (peningkatan polaritas) diperoleh fraksi C1 hingga C7. Diperoleh senyawa C6a, lalu diuji kemurnian dengan kromatografi lapis tipis dan HPLC. Senyawa hasil isolasi dianalisis dengan metode termal DSC, spektrometer FT-IR, spektrometer ¹H-NMR (Nuclear Magnetic Resonance), dan spektrometer 13C NMR. Berdasarkan hasil identifikasi senyawa metabolit sekunder dari ekstrak metanol fraksi C6 didapatkan senyawa yang golongan flavonoid yaitu alpinetin. Kristal yang didapat sebanyak 450 mg berwarna kekuningan dengan bentuk seperti jarum

Boesenbergia Rotunda plant, also known as fingerroot, is one of the plants used as a traditional medicinal plant. Fingerroot comes from Southeast Asia, namely from Indonesia, Malaysia and Thailand. This plant is used traditionally to cure stomach ulcers and cough medicine. There are secondary metabolite compounds that have the potential to act as antioxidant, antibacterial, anti-inflammatory and anti-mutagenic agents. In this research, secondary metabolite compounds were isolated from the rhizomes of fingerroot from Ponorogo. The method used to isolate the compound was maceration extraction with methanol to produce 230,53 grams of thick extract and differentiation using column chromatography. Results from vacuum liquid chromatography (VLC) were eluted using n-Hexane, dichloromethane, ethyl acetate, and methanol. The results of the separation were fractions A, B, C, and D. Fraction C of 15 grams was separated again using a gravity chromatography column (GCC) eluted with n-Hexane: ethyl acetate (increased polarity) to obtain fractions C1 to C7. Compound C6a was obtained, then its purity was tested using thin layer chromatography and HPLC. The isolated compounds were analyzed using the DSC thermal method, FT-IR spectrometer, ¹H-NMR (Nuclear Magnetic Resonance) spectrometer, and 13C NMR spectrometer. Based on the results of the removal of secondary metabolite compounds from the methanol extract of the C6 fraction, a compound belonging to the flavonoid group, namely alpinetin was obtained. The crystals obtained were 450 mg pale yellow colored with a needle-like shape.
