Abstract
Melon (Cucumis melo L.) merupakan tanaman hortikultura anggota famili Cucurbitaceae. Melon menjadi model yang menarik untuk mempelajari karakter pematangan buah karena adanya genotip klimakterik dan non-klimakterik dalam satu spesies. Pendekatan molekuler untuk mengidentifikasi karakter pematangan klimakterik dan non-klimkaterik secara komersial untuk meregulasi kualitas dan umur simpan buah. Penelitian ini bertujuan untuk mengetahui kandungan pigmen (klorofil a, klorofil b, dan total klorofil) dari tiga aksesi melon pada satuan waktu tertentu, profil ekspresi gen terpaut biosintesis pada tiga kultivar melon selama perkembangan buah dan karakter morfologi buah terkait pematangan. Tiga kultivar yang menjadi objek analisis terdiri dari ‘Melona’, ‘GMP’, dan ‘Tacapa Gold’. Pada masing-masing kutivar, terdapat tiga kelompok analisis berdasarkan usia kematangan, yaitu 15 DAA (days after anthesis), 30 DAA, dan 55 DAA. Dua gen terpaut klorofil yang dikarakterisasi, yaitu CmCAO dan CmChlg. Kandungan pigmen diukur dengan metode spektrofotometer UV-VIS dengan panjang gelombang 190-800 nano meter (nm). Profil ekspresi gen dieksplorasi dengan metode RT-qPCR. Pendekatan analisis ekspresi gen relatif menggunakan metode 2ˉΔΔCt. Total klorofil a dan b tertinggi pada bagian mesokarp buah kultivar ‘Gama Melon Parfum’ pada ketiga usia kematangan yakni usia kematangan 15 DAA (594,4±1,31(mg/mL bb)), 30 DAA (358,07±0,5(mg/mL bb)) dan 55 DAA (213,13±0,6(mg/mL bb)). Usia kematangan buah 55 DAA memiliki ekspresi gen yang rendah pada gen CmChlg sedangkan CmCAO memiliki ekspresi gen yang masih tinggi pada usia 55 DAA pada semua jenis kultivar. Warna hijau yang dominan pada buah disebabkan oleh klorofil a. Eksokarp pada melon menunjukkan kandungan klorofil a yang lebih tinggi dan klorofil b daripada mesokarp.

Melon (Cucumis melo L.) is a horticultural plant belonging to the Cucurbitaceae family. Melon is an interesting model for studying fruit ripening characteristics because of the existence of climacteric and non-climacteric genotypes within one species. Molecular approaches to identify climacteric and non-climacteric ripening traits commercially to regulate fruit quality and shelf life. This study aims to determine the pigment content (chlorophyll a, chlorophyll b, and total chlorophyll) of three melon accessions at a certain time, the expression profile of biosynthesis-related genes in three melon cultivars during fruit development and fruit morphological characters related to ripening. The three cultivars that were the object of analysis consisted of 'Melona', 'GMP', and 'Tacapa Gold'. For each cultivar, there were three analysis groups based on age of maturity, namely 15 DAA (days after anthesis), 30 DAA, and 55 DAA. Two chlorophyll-linked genes were characterized, namely CmCAO and CmChlg. Pigment content was measured using the UV-VIS spectrophotometer method with a wavelength of 190-800 nano meters (nm). Gene expression profiles were explored using the RT-qPCR method. The relative gene expression analysis approach uses the 2ˉΔΔCt method. The highest total chlorophyll a and b were in the mesocarp of the cultivar 'Gama Melon Parfum' at the three ages of maturity, namely 15 DAA (594,4±1,31(mg/mL bw)), 30 DAA (358,07±0,5(mg/mL bw)) and 55 DAA (213,13±0,6(mg/mL bw)). The fruit maturity age of 55 DAA has low gene expression of the CmChlg gene while CmCAO has high gene expression at the age of 55 DAA in all types of cultivars. The dominant green color in fruit is caused by chlorophyll a. The exocarp of melon shows a higher content of chlorophyll a and chlorophyll b than the mesocarp.
