Abstract
Isolasi rimpang temu kunci dilakukan untuk mengambil senyawa panduratin A dari temu kunci. Isolasi dilakukan dengan metode ekstraksi menggunakan maserasi dengan pelarut metanol pada suhu ruang dan dilanjutkan fraksinasi menggunakan kromatografi cair vakum dengan kontrol pemisahan menggunakan KLT. Hasilnya dari ekstrak metanol yang dikromatografi cair vakum menghasilkan empat fraksi dan dinamai fraksi A, B, C dan D. Fraksi B difraksinasi menggunakan KCV menghasilkan delapan fraksi B1-B8. Dari fraksi B3 didapatkan padatan kuning dengan titik leleh 157,3°C. Hasil uji KLT tiga eluen, KLT 2D dan KCKT menunjukkan padatan kuning yang diambil dari fraksi B3 adalah senyawa tunggal dan dinamai senyawa B3. Penentuan struktur senyawa B3 dilakukan menggunakan Spektrofotometer IR untuk mengetahui gugus fungsi senyawa B3 dan hasilnya terdapat ikatan karbonil, hidroksil serta ikatan karbon-oksigen. Selanjutnya untuk mengetahui profil proton dan karbon dianalisis menggunakan RMI proton dan karbon. Berdasarkan analisis elusidasi struktur menggunakan spektrofotometer IR, RMI proton dan karbon didapatkan bahwa senyawa B3 adalah senyawa panduratin A

Isolation of ginger rhizomes was carried out to extract the panduratin A compound from fingerroot. Isolation was carried out using an extraction method using maceration with methanol solvent at room temperature and continued with fractionation using vacuum liquid chromatography with inspection control using TLC. The results of the methanol extract which were subjected to vacuum liquid chromatography produced four fractions and were named fractions A, B, C and D. Fraction B which was fractionated using VLC produced eight fractions B1-B8. From fraction B3, a yellow solid was obtained with a melting point of 157.3°C. The results of the three eluent TLC, 2D TLC and HPLC tests showed that the yellow solid taken from the B3 fraction was a single compound and was named compound B3. The structure of the B3 compound was determined using an IR Spectrophotometer to determine the functional groups of the B3 compound and the results showed that there were carbonyl, hydroxyl and carbon-oxygen bonds. Next, to find out the proton and carbon profiles, they were analyzed using RMI protons and carbon. Based on structural elucidation analysis using an FTIR spectrophotometer, proton and carbon NMR, it was found that B3 compound is a panduratin A compound.
