Abstract

Mikroalga merupakan organisme fotoautotrof yang memiliki kemampuan menghasilkan berbagai senyawa bioaktif termasuk antioksidan. Tujuan dari penelitian ini adalah untuk melakukan identifikasi dan uji kapasitas antioksidan isolat mikroalga yang berhasil dikultivasi dari perairan Kolam Asrama Mahasiswa ITS. Isolasi mikroalga dilakukan dengan metode pengenceran bertingkat dan streak plate pada sampel air. Isolat yang diidentifikasi secara morfologi dan molekuler (analisis sekuens gen 18S rRNA dan pohon filogenetik) selanjutnya diuji kapasitas antioksidannya. Uji kapasitas antioksidan dilakukan dengan menggunakan metode DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2’-azino-bis(3-etilbenzotiazolin-6-sulfonat)), dan FRAP (Ferric Reducing Antioxidant Power) berdasarkan hasil analisis nilai IC50 yang didapatkan. Isolat mikroalga yang berhasil dikultivasi dan diidentifikasi secara morfologi dan molekuler antara lain Chlorella vulgaris, Vitreochlamys incisa, Dictyosphaerium ehrenbergianum, dan Desmodesmus armatus. Kekuatan nilai IC50 tertinggi pada metode ABTS didapatkan dari ekstrak metanol isolat KD3a (Chlorella vulgaris) dengan karakteristik antioksidan kuat (81,693 ppm) dan nilai IC50 terendah didapatkan dari ekstrak etanol isolat 17HtDB2b (Desmodesmus armatus) (87278,031 ppm / 87,2 mg/mL). Untuk Uji DPPH, kekuatan nilai IC50 tertinggi ada pada ekstrak metanol isolat KD3a (297,451 ppm) dan nilai IC50 terendah didapatkan dari ekstrak metanol isolat 17HtDB2b (285213,013 ppm / 285,2 mg/mL). Pada uji FRAP didapatkan nilai tertinggi pada ekstrak etanol isolat HDB3a (Dictyosphaerium ehrenbergianum) (74.45 mg AAE/g) yang memiliki karakteristik antioksidan baik dan terendah pada isolat 17HtDB2b (5,58 mg AAE/g).

Microalgae are photoautotrophic organisms with the capability to produce various bioactive compounds, including antioxidants. This study aims to identify and evaluate the antioxidant capacity of microalgal isolates successfully cultivated from the ITS Student Dormitory pond. Microalgae were isolated using a dilution series and streak plate methods on water samples. The isolates were identified through morphological and molecular analyses (18S rRNA gene sequencing and phylogenetic tree construction) and subsequently tested for antioxidant capacity. Antioxidant capacity was assessed using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)), and FRAP (Ferric Reducing Antioxidant Power) assays based on the IC50 values obtained. The microalgal isolates identified morphologically and molecularly include Chlorella vulgaris, Vitreochlamys incisa, Dictyosphaerium ehrenbergianum, and Desmodesmus armatus. The highest IC50 value in the ABTS assay was observed for the methanol extract of isolate KD3a (Chlorella vulgaris) with strong antioxidant characteristics (81.693 ppm), while the lowest IC50 value was obtained from the ethanol extract of isolate 17HtDB2b (Desmodesmus armatus) (87,278.031 ppm / 87.2 mg/mL). In the DPPH assay, the highest IC50 value was recorded for the methanol extract of isolate KD3a (297.451 ppm), and the lowest IC50 value was observed for the methanol extract of isolate 17HtDB2b (285,213.013 ppm / 285.2 mg/mL). The FRAP assay revealed the highest value in the ethanol extract of isolate HDB3a (Dictyosphaerium ehrenbergianum) (74.45 mg AAE/g), indicating good antioxidant characteristics, and the lowest value in isolate 17HtDB2b (5.58 mg AAE/g).
