Abstract

Scaffold bone graft adalah struktur biomaterial 3 dimensi yang digunakan sebagai salah satu solusi rekontruksi cacat tulang. Material yang dipilih pada penelitian ini adalah hidroksiapatit (HAp) yang diperoleh dari sintesis tulang sapi menggunakan bantuan microwave, oligokitosan (Cs) dan poly(methyl methacrylate) (PMMA). Penelitian ini bertujuan untuk menganalisis efek rasio komposisi terhadap sifat mekanik dan biokompatibilitas scaffold. Metode yang digunakan untuk fabrikasi scaffold adalah solvent casting/particulate leaching (SCPL) dengan menggunakan NaCl sebagai porogen dan glutaraldehid sebagai crosslinker. Variasi rasio komposisi untuk HAp:Cs:PMMA yaitu, 68:2:30; 66:4:30 dan 64:6:30. Pengujian yang dilakukan berupa analisis morfologi menggunakan stereo mikroskop yang menunjukkan terbentuknya interconnected porous yang baik. Hasil analisis FTIR pada scaffold yang berhasil difabrikasi didapat gugus fungsi yaitu peak O-H stretching pada 3550 sampai 3050 cm-1, kemudian C-H stretching pada 2961, 2916 dan 2848 cm-1, C=O stretching pada 1688 cm-1, amida I pada 1653 cm-1, amida II pada 1517 cm-1, amida III pada 1369 cm-1, C-N stretching pada 1096 cm-1, C=N/imine bond pada 1542 cm-1. Kemudian gugus karbonat, fosfat dan hidroksil pada 1411 dan 1467 cm-1; 1015 cm-1 dan 711 cm-1. Perhitungan menggunakan liquid displacement test menunjukkan nilai porositas tertinggi terdapat pada sampel Cs6HAp64 yaitu 87,92%, pengujian anti bakteri mendapatkan zona hambat terbesar pada sampel Cs6HAp64 yaitu sebesar 13,67 mm, porositas dan sifat anti bakteri dipengaruhi oleh penambahan kandungan kitosan yang bervariasi. Penambahan hidroksiapatit dan PMMA berpengaruh pada sifat mekanik yaitu nilai kuat tekan tertinggi pada sampel Cs2HAp68 sebesar 30,35 MPa dan terendah pada sampel Cs6HAp64 sebesar 10,16 MPa. Pengujian MTT Assay menunjukkan hasil viabilitas sel tertinggi pada sampel Cs6HAp64 sebesar 75,77% angka ini masuk dalam kategori slightly cytotoxic.

composition ratio on the mechanical properties and biocompatibility of the scaffold. The method used for the scaffold fabrication is solvent casting/particulate leaching (SCPL) using NaCl as a porogen and glutaraldehyde as a crosslinker. The composition ratio for HAp:Cs:PMMA were, 68:2:30; 66:4:30 and 64:6:30. The tests carried out were in the form of
morphological analysis using a stereo microscope which showed good interconnected porous formation. The results of the FTIR analysis on the successfully fabricated scaffold obtained functional groups, namely peak O-H stretching at 3550 to 3050 cm-1, then C-H stretching at 2961, 2916 and 2848 cm-1, C=O stretching at 1688 cm-1, amide I at 1653 cm-1
, amide II at 1517 cm-1, amide III at 1369 cm-1, C-N stretching at 1096 cm-1, C=N/imine bond at 1542 cm-1. Then the carbonate, phosphate and hydroxyl groups at 1411 and 1467 cm-1; 1015 cm-1and 711 cm-1. Calculations using the liquid displacement test showed that the highest porosity value was found in the Cs6HAp64 sample, which was 87.92%, the antibacterial test got the largest inhibition zone in the Cs6HAp64 sample, which was 13.67 mm, the porosity and antibacterial properties were influenced by the addition of varying concentrations of chitosan. The addition of hydroxyapatite and PMMA has an effect on the mechanical properties, namely the highest compressive strength value in the Cs2HAp68 sample of 30.35 MPa and the lowest in the Cs6HAp64 sample of 10.16 MPa. The MTT Assay test showed the highest cell viability results in the Cs6HAp64 sample of 75.77%, this number was in the slightly cytotoxic category.
