Abstract

Praktik Keinsinyuran ini bertujuan untuk mengaplikasikan teknologi ultrafiltrasi two-stage dan penambahan maltodekstrin sebagai protektan dalam produksi serbuk enzim bromelin dari bonggol nanas. Topik ini dipilih untuk menjawab kebutuhan industri akan teknologi pemurnian enzim yang lebih efisien dan ramah lingkungan, serta untuk mendukung pemanfaatan limbah pertanian sebagai sumber daya bernilai tinggi. Sebagai bagian dari Praktik Keinsinyuran, penerapan teknologi ini tidak hanya menguji aspek teknis dari proses, tetapi juga melibatkan evaluasi manajemen sumber daya, penjadwalan, dan kontrol kualitas dalam proses produksi. Teknologi ultrafiltrasi two-stage dipilih karena kemampuannya dalam memurnikan enzim secara efektif, memisahkan kontaminan dengan hasil pemurnian yang lebih tinggi dibandingkan metode konvensional, dan lebih hemat energi. Selain itu, penambahan maltodekstrin digunakan untuk melindungi enzim selama proses pengeringan dan memastikan aktivitas serta stabilitas produk akhir tetap optimal. Kegiatan Praktik Keinsinyuran ini mencakup analisis manajemen proses produksi, optimalisasi parameter teknis, serta evaluasi penerapan teknologi dalam skala industri. Hasil kegiatan menunjukkan bahwa penggunaan ultrafiltrasi two-stage mampu meningkatkan pemurnian enzim hingga 93,3%, dengan aktivitas spesifik enzim mencapai 61.333 U/g, lebih tinggi dari standar komersial. Penambahan maltodekstrin sebesar 6% menghasilkan serbuk enzim yang stabil dengan kadar protein 4,5 mg/mL. Analisis gugus fungsi melalui FTIR menunjukkan kesamaan antara enzim bromelin yang dihasilkan dengan produk komersial yang ada di pasar.

This Engineering Practice aims to apply two-stage ultrafiltration technology and the addition of maltodextrin as a protectant in the production of bromelain enzyme powder from pineapple pith. This topic was chosen to answer the industry's need for a more efficient and environmentally friendly enzyme purification technology, as well as to support the utilization of agricultural waste as a high value resource. As part of the Engineering Practice, the application of this technology not only tests the technical aspects of the process, but also involves the evaluation of resource management, scheduling, and quality control in the production process. The two-stage ultrafiltration technology was chosen due to its ability to effectively purify enzymes, separate contaminants with higher purification yields than conventional methods, and is more energy efficient. In addition, the addition of maltodextrin is used to protect the enzymes during the drying process and ensure the activity and stability of the final product remain optimal. This Engineering Practice activity includes analysis of production process management, optimization of technical parameters, and evaluation of technology application on an industrial scale. The results showed that the use of two-stage ultrafiltration was able to increase enzyme purification to 93.3%, with enzyme specific activity reaching 61,333 U/g, higher than commercial standards. The addition of 6% maltodextrin produced a stable enzyme powder with a protein content of 4.5 mg/mL. Functional group analysis through FTIR showed similarities between the bromelain enzyme produced and commercial products on the market.
