Abstract
Antioksidan adalah senyawa yang mampu menghambat, mencegah, dan mengurangi aktivitas radikal bebas. Antioksidan berperan penting dalam melindungi sel-sel tubuh dari kerusakan akibat ROS dan kerusakan DNA/protein. Buah delima atau pomegranate (Punica granatum L.) telah banyak dimanfaatkan dalam bidang pangan dan kesehatan karena memiliki sifat antioksidan, antimikroba dan therapeutic. Penelitian bertujuan untuk menganalisis kapasitas antioksidan ekstrak n-heksana eksokarp dan mesokarp tiga varietas buah delima (Punica granatum L.) yaitu delima merah, delima putih, dan delima ungu, yang memiliki warna kulit berbeda. Sampel yang digunakan dalam penelitian ini adalah eksokarp dan mesokarp dari tiga varietas delima yang diekstraksi menggunakan pelarut n-heksana dengan analisis yang dilakukan secara terpisah untuk masing-masing bagian. Kapasitas antioksidan diukur dengan metode Total Phenol Compound (TPC), Total Flavonoid Compound (TFC), ABTS (2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid) dan DPPH (2,2-diphenyl-1-picrylhydrazyl) secara kolorimetri menggunakan spektrofotometer. Hasil penelitian menunjukkan TPC tertinggi ditemukan pada mesokarp delima merah (70,21 mgGAE/g), diikuti mesokarp delima putih (23,57 mgGAE/g), mesokarp delima ungu (17,71 mgGAE/g), eksokarp delima putih (2,60 mgGAE/g), eksokarp delima ungu (1,88 mgGAE/g), eksokarp delima merah (1,64 mgGAE/g). Sementara itu, kandungan TFC tertinggi terdapat pada mesokarp delima merah (12,07 mgQE/g), diikuti mesokarp delima putih (6,60 mgQE/g), mesokarp delima ungu (3,86 mgQE/g), eksokarp delima putih (1,98 mgQE/g), eksokarp delima ungu (1,30 mgQE/g), eksokarp delima merah (0,96 mgQE/g). Uji kapasitas antioksidan menunjukkan bahwa ekstrak n-heksana mesokarp buah delima memiliki kapasitas antioksidan yang lebih tinggi dibandingkan eksokarp buah delima. Kapasitas antioksidan tertinggi ditemukan pada mesokarp buah delima merah dengan nilai IC50 pada uji ABTS 234,00 μg/mL dan 423,79 μg/mL pada uji DPPH. Kapasitas antioksidan baik pada eksokarp dan mesokarp delima termasuk dalam kategori antioksidan sangat lemah dengan IC50 >200 ppm. Hasil penelitian ini menjelaskan terkait kapasitas antioksidan dari bagian eksokarp dan mesokarp tiga varietas delima dan dapat digunakan sebagai informasi awal sumber baru antioksidan.

Antioxidants are compounds that can inhibit, prevent, and reduce free radical activity. They play a crucial role in protecting body cells from damage caused by reactive oxygen species (ROS) and DNA/protein damage. Pomegranate (Punica granatum L.) has been widely utilized in the food and health sectors due to its antioxidant, antimicrobial, and therapeutic properties. This study aimed to analyze the antioxidant capacity of n-hexane extracts from the exocarp and mesocarp of three pomegranate varieties (red, white, and purple), characterized by their different peel colors. The samples used in this research consisted of the exocarp and mesocarp of the three pomegranate varieties, which were extracted using n-hexane, with separate analyses performed for each part. The antioxidant capacity was measured using Total Phenolic Compound (TPC), Total Flavonoid Compound (TFC), ABTS (2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid), and DPPH (2,2-diphenyl-1-picrylhydrazyl) assays via colorimetric methods using a spectrophotometer. The results revealed that the highest TPC was observed in the mesocarp of the red pomegranate (70,21 mgGAE/g), followed by the mesocarp of the white pomegranate (23,57 mgGAE/g), the mesocarp of the purple pomegranate (17,71 mgGAE/g), the exocarp of the white pomegranate (2,59 mgGAE/g), the exocarp of the purple pomegranate (1,88 mgGAE/g), and the exocarp of the red pomegranate (1,64 mgGAE/g). Similarly, the highest TFC was found in the mesocarp of the red pomegranate (12,07 mgQE/g), followed by the mesocarp of the white pomegranate (6,60 mgQE/g), the mesocarp of the purple pomegranate (3,86 mgQE/g), the exocarp of the white pomegranate (1.98 mgQE/g), the exocarp of the purple pomegranate (1,30 mgQE/g), and the exocarp of the red pomegranate (0,96 mgQE/g). Antioxidant capacity assays indicated that the n-hexane extract of the mesocarp exhibited higher antioxidant capacity compared to the exocarp. The highest antioxidant capacity is observed in the mesocarp of the red pomegranate, as evidenced by IC50 values of 234,00 μg/mL (ABTS assay) and 423,79 μg/mL (DPPH assay). Both the exocarp and mesocarp of the pomegranate demonstrated very weak antioxidant activity, as indicated by IC50 values exceeding 200 ppm. This study provides insights into the antioxidant capacity of the exocarp and mesocarp of three pomegranate varieties, which may serve as preliminary information on new potential sources of antioxidants.
