Abstract
Metil oranye (MO) merupakan salah satu zat warna yang banyak digunakan dalam industri tekstil. Namun, tingginya tingkat penggunaan MO tidak sebanding dengan pengelolaan limbah yang ditimbulkan karena sifat toksisitas dan karsinogeniknya. Tujuan penelitian ini adalah menyelidiki biodegradasi pewarna MO dengan bakteri Ralstonia pickettii yang terimobilisasi dalam matriks sodium alginate/polivinil alkohol/arang aktif (SA/PVA/AA). Beads dibuat dengan mencampurkan SA, PVA, dan AA dalam 50 mL aqua DM steril, dengan komposisi maisng-masing 2:4:1 (% b/v). Variasi beads yang dibuat yaitu SA/PVA/AA/Bakteri hidup, SA/PVA/AA/Bakteri mati, dan SA/PVA/AA tanpa bakteri. Dekolorisasi dilakukan dengan menginokulasikan beads dalam 100 mL larutan MO 50 mg/L selama 10 hari inkubasi dengan suhu 37 °C. Ketiga jenis beads menghasilkan persen dekolorisasi masing-masing sebesar 87,39% untuk SA/PVA/AA/Bakteri hidup; 38,76% untuk SA/PVA/AA/Bakteri mati; dan 36,99% untuk SA/PVA/AA . Dekolorisasi oleh free cell (FC) bakteri juga dilakukan sebagai perbandingan, dimana FC hidup sebesar 15,74% dan FC mati sebesar 10,64%. Hasil dekolorisasi beads jauh lebih baik dibanding dekolorisasi free cell bakteri. Keberhasilan imobilisasi untuk biodegradasi MO didukung dengan spektra FTIR dan visualisasi SEM. Uji reusabilitas beads dilakukan sebanyak tiga kali siklus, dengan persen dekolorisasi sebesar 87,39%; 46,05%; dan 31,42%. Hasil ini menunjukkan bahwa bakteri R. pickettii yang terimobilisasi beads SA/PVA/AA dapat digunakan untuk dekolorisasi MO dan dijadikan alternatif untuk mengurangi permasalahan limbah pewarna tekstil yang ada.===================================================================================================================================
Methyl orange (MO) is a dye that is widely used in the textile industry. However, the high level of use of MO is not commensurate with the management of the waste generated due to its toxicity and carcinogenic properties. The aim of this study was to investigate the biodegradation of MO dye with Ralstonia pickettii bacterium immobilized in sodium alginate/polyvinyl alcohol/activated carbon (SA/PVA/AA) matrix. Beads were made by mixing SA, PVA, and AA in 50 mL sterile aqua DM, with the compositions being 2:4:1 (%w/v). The variations of beads formed were SA/PVA/AA/live bacteria, SA/PVA/AA/dead bacteria, and SA/PVA/AA without bacteria. Decolorization was carried out by inoculating the beads in 100 mL of 50 mg/L MO solution for 10 days of incubation at 37 °C. The three types of beads resulted in the percentage of decolorization by 87.39% for SA/PVA/AA/live bacteria; 38.76% for SA/PVA/AA/dead bacteria; and 36.99% for SA/PVA/AA without bacteria. Decolorization by free cell (FC) bacteria was also carried out as a comparison, with the results by 15.74% for live FC bacteria and 10.64% for dead FC bacteria. The result of MO decolorization by immobilized beads was much better than that by bacterial free cell. The success of immobilization for MO biodegradation was supported by FTIR spectra and SEM visualization. The beads reusability test was carried out in three cycles, with the percent decolorization of 87.39%; 46.05%; and 31.42%. These results indicated that the immobilized R. pickettii bacterium in SA/PVA/AA matrix can be used for MO decolorization and can be used as an alternative to reduce the existing textile dye waste problem.
