Abstract
Sungai brantas merupakan perairan baku mutu III dan memiliki keanekaragaman hayati yang belum terdokumentasi secara menyeluruh. Salah satu pendekatan yg digunakan adalah metode eDNA metabarcoding, dan bisa menggunakan Spons sebagai sumber eDNA. Spons bersifat non-selective filter feeder sehingga berpotensi digunakan sebagai bioindikator melalui pendekatan eDNA metabarcoding. Penelitian ini kami akan
menggunakan eDNA spons dan membandingkannya dengan sampel eDNA air. Tujuan Penelitian ini mampu membantu mengetahui keanekaragaman hayati dan mengetahui perbandingan keanekaragaman hayati yang ditemukan dari sampel yang diambil pada Sungai Brantas, Mojokerto. Hasil penelitian menunjukkan bahwa ketiga jenis sampel air, spons Eunapius carteri, dan spons Oncosclera asiatica memiliki tingkat keanekaragaman hayati (Alpha Diversity) yang tinggi, dengan nilai indeks >3. Indeks Jaccard menunjukkan tingkat kesamaan komunitas (Beta Diversity) antar sampel berada dalam kategori sedang (15%–50%). Sampel air memiliki jumlah taksa unik terbanyak, baik pada tingkat kelas (51 taksa) maupun genus (792 taksa). Temuan ini menunjukkan bahwa meskipun spons air tawar mampu merekam biodiversitas dengan baik melalui eDNA, sampel air memberikan cakupan taksonomi yang lebih luas dan representatif. Oleh karena itu, air dinilai sebagai media pengambilan sampel yang lebih optimal untuk menggambarkan struktur komunitas di perairan, karena air sebagai habitat seluruh makhluk hidup dan mikroorganisme.

The Brantas River is a Class III water body and has biodiversity that has not been fully documented. One of the approaches used is the eDNA metabarcoding method, and sponges can be used as a source of eDNA. Sponges are non-selective filter feeders and thus have the potential to be used as bioindicators through the eDNA metabarcoding approach. In this study, we will use sponge eDNA and compare it with water eDNA samples. The objective of this study is to help determine biodiversity and compare the biodiversity found in samples taken from the Brantas River in Mojokerto. The results showed that all three types of water samples, Eunapius carteri sponges, and Oncosclera asiatica sponges had high levels of biodiversity (Alpha Diversity), with an index value >3. The Jaccard index indicated that the similarity of communities (Beta Diversity) between samples was in the moderate category (15%–50%). Water samples had the highest number of unique taxa, both at the class level
(51 taxa) and genus level (792 taxa). These findings indicate that while freshwater sponges can effectively record biodiversity through eDNA, water samples provide a broader and more representative taxonomic coverage. Therefore, water is considered a more optimal sampling medium for describing community structure in aquatic environments, as water serves as the habitat for all living organisms and microorganisms.
