Abstract
Ganoderma boninense merupakan patogen primer yang menyebabkan busuk pangkal batang (BPB) pada kelapa sawit. Saat ini, upaya pengendalian jamur telah dilakukan dengan menggunakan metode fisik, agen biologi, dan senyawa kimia, namun belum berhasil sepenuhnya dalam membunuh jamur. Penelitian ini merupakan suatu upaya inovasi pengendalian jamur dengan mengkombinasikan gelombang ultrasonik, kitosan, dan ultraviolet-C secara berurutan untuk mengetahui pengaruhnya terhadap pertumbuhan G. boninense. Selama 14 hari dilakukan pengamatan dan pengukuran diameter koloni dengan aplikasi paparan dilakukan sebanyak 2 tahap aplikasi. Aplikasi dilakukan secara berurutan dari gelombang ultrasonik, kitosan, dan ultraviolet-C pada hari yang sama. Hasil pengamatan diameter koloni pada pengaruh 2 kombinasi yaitu ultrasonik + UV-C, ultrasonik + kitosan, dan kitosan + UV-C belum cukup untuk menekan pertumbuhan jamur. Sedangkan pada perlakuan pengaruh tiga kombinasi, terdapat kelompok isolat yaitu ABC (ultrasonik 5’kHz + kitosan 0,5% + UV-C 100s) yang mengalami penghambatan pertumbuhan dengan diameter akhir sebesar 13,927±0,68 mm dan kematian yang diamati dari perubahan morfologi miselium yang lebih tipis dan terjadi perubahan warna. Perhitungan PIRG pada isolat ABC mencapai 84,52% yang dikategorikan sebagai “high”. Pengamatan morfologi hifa melalui scanning electron microscope didapatkan bahwa hifa mengalami abnormalitas yang memipih, menyatu, membengkak, mengerut kolaps, patah, dan berukuran abnormal. Perlakuan ABC diketahui berpengaruh terhadap pertumbuhan G. boninense.

Ganoderma boninense is a primary pathogen that causes basal stem rot (BSR) in oil palm. Currently, efforts to control the fungus have been made using physical methods, biological agents, and chemical compounds, but have not been fully successful in killing the fungus. This study aims to innovate fungal control by combining ultrasonic waves, chitosan, and ultraviolet-C sequentially to determine its effect on the growth of G. boninense. Within 14 days, observations and measurements of colony diameter were made with 2 stages of exposure application. Application is performed sequentially from ultrasonic waves, chitosan, and ultraviolet-C on the same day. The observation of colony diameter in the effect using 2 combinations are ultrasonic + UV-C, ultrasonic + chitosan, and chitosan + UV-C does not sufficient in suppressing the fungal growth. While in the treatment of the effect using three combinations the isolate ABC group (ultrasonic 40 kHz for 5 minutes + chitosan 0.5% + UV-C 100s) sufficient in surpressing the growth inhibition even killing the fungus with a final diameter of 13.927 ± 0.68 mm and change in mycelium morphological which is thinner and discolored. PIRG calculation on ABC isolate reached 84.52% which is categorized as “high”. Observation of hyphae morphology using scanning electron microscope found that hyphae undergoes abnormalities that were flattened, fused, swollen, shrunken collapsed, broken, and abnormal size. ABC treatment is known to affect the growth of G. boninense.
