Abstract
Alginat merupakan polisakarida alami yang banyak ditemukan pada alga coklat, khususnya Sargassum sp, yang dapat diaplikasikan di industri makanan, farmasi hingga kosmetik. Proses ekstraksi yang dilakukan secara konvensional memerlukan waktu yang lebih lama dan mendapatkan hasil yang kurang efektif. Penelitian ini bertujuan untuk melihat potensi penggunaaan ultrasonik sebagai metode pada proses ekstraksi alginat dengan jalur sodium agar lebih efisien dalam proses ekstraksi. Proses ekstraksi dilakukan dengan tiga tahap utama yaitu pre-extraction, neutralization, dan purification/precipitation. Penelitian ini menerapkan variasi molaritas Na2CO3 (0.1, 0.2, dan 0.3M) dengan waktu ekstraksi (60, 120, dan 180 menit). Pengujian dilakukan untuk mengevaluasi kualitas hasil ekstraksi alginat dengan berdasarkan standar ASTM 2064 yang menjelaskan mengenai alginat untuk biomedis. Didapatkan nilai rendemen terbesar pada sampel dengan molaritas Na2CO3 0.2 molar dan variasi waktu 120 menit yaitu sebesar 16.13%. Pada pengujian kadar air didapatkan nilai masuk dalam standar kadar air dibawah 15% dengan sampel paling tinggi ada pada sampel M0.3W120 dengan nilai 13% dan paling kecil ada pada sampel M0.1W120 dengan nilai 9.2%. Pada pengujian GPC menentukan nilai berat molekul sampel, semakin tinggi waktu ekstraksi maka nilai berat molekulnya semakin turun, begitu juga pada konsentrasi Na2CO3. Pada pengujian viskositas, didapatkan nilai yang sejalan dengan pengujian GPC semakin tinggi konsentrasi Na2CO3, maka akan semakin rendah viskositasnya. Variasi waktu juga berpegaruh terhadap nilai viskositasnya, semakin tinggi waktu ekstraksinya maka semakin rendah nilai viskositasnya. Pada pengujian NMR digunakan untuk melihat rasio M/G sampel ini yaitu 0.460 dengan blok G; blok M yaitu 0.685; 0.315.

Alginate are natural polysaccharides commonly found in brown algae, particularly Sargassum sp., and can be applied in industries such as food, pharmaceuticals, and cosmetics. Conventional extraction processes require longer times and yield less effective results. This study aims to explore the potential use of ultrasonic waves as a method in the extraction process of alginate with sodium, to make the extraction process more efficient. The extraction process was carried out in three main stages: pre-extraction, neutralization, and purification/precipitation. This study applied variations in the molarity of Na₂CO₃ (0.1, 0.2, and 0.3 molar) with extraction times (60, 120, and 180 minutes). The tests were conducted to evaluate the quality of the alginate extract based on the ASTM 2064 standard, which describes alginate for biomedical use. The highest yield was obtained from the sample with 0.2 M Na₂CO₃ and an extraction time of 120 minutes, yielding 16.13%. In the moisture content test, the values were found to meet the standard moisture content of below 15%, with the highest moisture content in the sample M0.3W120 at 13%, and the lowest in the sample M0.1W120 at 9.2%. The GPC test determined the molecular weight of the sample, showing that the longer the extraction time, the lower the molecular weight, which also correlates with the concentration of Na₂CO₃. In the viscosity test, the results aligned with the GPC test, where higher Na₂CO₃ concentrations led to lower viscosity values. The variation in extraction time also influenced the viscosity, with longer extraction times resulting in lower viscosity. The NMR test was used to determine the M/G ratio of this sample, which was 0.460 with the G block at 0.685 and the M block at 0.315.
