Abstract
Pule pandak (Rauvolfia serpentina (L.)) merupakan tanaman herbal yang memiliki manfaat farmakologis karena kandungan metabolit sekunder berupa alkaloid, namun teknik budidaya konvensional memiliki banyak kelemahan sehingga diperlukan metode alternatif berupa kultur in vitro yang cepat, aseptik, dan terkontrol. Peningkatan kandungan metabolit sekunder dapat dilakukan melalui teknik elisitasi menggunakan melatonin yang dapat meningkatkan pertumbuhan dan sintesis metabolit sekunder R. serpentina. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh dari aplikasi melatonin eksogen terhadap pertumbuhan dan biosintesis metabolit sekunder pada pule pandak (R. serpentina) secara in vitro. Penelitian dilakukan dengan penanaman eksplan R. serpentina secara in vitro pada media kultur dengan perlakuan melatonin 0 µM, 1 µM, 5 µM, dan 10 µM. Parameter yang diamati berupa persentase induksi organogenesis, kalogenesis, tinggi tanaman, berat basah, jumlah tunas, daun, nodus, serta profil metabolit dengan untargeted metabolomics uji LC-HRMS. Hasil menunjukkan seluruh perlakuan memiliki persentase organogenesis tunas sebesar 100 % dan akar sebesar 0 %, dan kalus hanya terbentuk pada perlakuan 1 µM dengan persentase 83,3 %.
Parameter pertumbuhan ditunjukkan paling optimal pada 1 µM dengan peningkatan jumlah daun, nodus, dan tinggi tanaman. Profil metabolit menunjukkan jumlah senyawa tertinggi sebesar 1421 pada perlakuan 1 dan 5 µM. Analisis multivariasi PCA, PLS-DA, dan HCA 100 metabolit menunjukkan golongan metabolit tertinggi pada alkaloid sebesar 51 dan membentuk 2 kluster yakni kluster 0 µM dan 1 µM, serta kluster 5 µM dengan 10 µM. Metabolit sekunder yang terdeteksi sebagai senyawa VIP akibat pengaruh pemberian melatonin antara lain dari golongan alkaloid, fenol, flavonoid, sterol, steroid, poliketida, quinone, dan karotenoid.

Pule pandak (Rauvolfia serpentina (L.)) is a herbal plant with pharmacological benefits due to its secondary metabolite content in the form of alkaloids. However, conventional cultivation techniques have many shortcomings, necessitating an alternative method in the form of rapid, aseptic, and controlled in vitro culture. The content of secondary metabolites can be increased through elicitation techniques using melatonin, which can enhance growth and the synthesis of secondary metabolites in R. serpentina. The objective of this study was to investigate the effect of exogenous melatonin application on growth and the biosynthesis of secondary metabolites in Pule Pandak (R. serpentina) in vitro. The study was conducted by culturing R. serpentina explants in vitro on culture media with melatonin treatments of 0 µM, 1 µM, 5 µM, and 10 µM. The parameters observed included the percentage of organogenesis induction, callogenesis, plant height, fresh weight, number of shoots, leaves, nodes, and metabolite profiles using untargeted metabolomics LC-HRMS analysis. The results showed that all treatments had a 100% organogenesis rate for shoots and 0% for roots, and callus formation only occurred in the 1 µM treatment with an 83.3% rate. Growth parameters were most optimal at 1 µM, with increases in the number of leaves, nodes, and plant height. The metabolite profile showed the highest number of compounds (1,421) in the 1 and 5 µM treatments. Multivariate analysis using PCA, PLS-DA, and HCA of 100 metabolites indicated that higher group contains from alkaloid group with 51 metabolites and clustered to 2 groups 0 and 1 µM, 5 µM and 10 µM. Secondary metabolites detected as VIP compounds due to melatonin administration include alkaloids, phenols, flavonoids, sterols, steroids, polyketides, quinones, and carotenoids.
