Abstract

Kurma (Phoenix dactylifera) merupakan tumbuhan yang tergolong dalam family Arecaceae. Kurna memiliki kematangan reproduksi biasanya antara 4-5 tahun sehingga pada tahap awal pembibitan tidak dapat ditentukan jenis kelamin jantan atau betina. Tujuan dari penelitian ini yaitu mendeterminasi jenis kelamin Kurma melalui karakteristik fenotip dan molekuler gen SRY yang divalidasi menggunakan ISSR Marker. Sampel Kurma yang digunakan sebanyak 10 bibit dari masing-masing kultivar yaitu KL.1 dan Barhe. Metode penelitian ini meliputi perbanyakan Kurma secara In Vivo sehingga dapat dikarakterisasi secara fenotip melalui struktur radikula (berulir atau tidak berulir) dan struktur daun primordia (melengkung atau tidak melengkung). Determinasi jenis kelamin jantan dan betina juga dilakukan secara molekuler dengan menggunakan marka molekuler Gen SRY (SRY-date F-R) dan ISSR Marker (IS_A02 dan IS_A71). Hasil karakterisasi fenotip Kurma kultivar KL. 1 menghasilkan 7 bibit jantan (K1, K3, K4, K5, K12, K14, K15) dan 3 betina (K2, K6, K9). Sementara itu, Kurma kultivar Barhe terindikasi 7 jantan yaitu bibit B1, B2, B5, B8, B11, B12, B15 dan 3 betina yaitu B4, B6, B10. Selain itu, dilakukan determinasi secara molekuler dimana hasil pada kultivar KL 1 telah memvalidasi karakterisasi fenotip dengan indikasi jenis kelamin bibit yang sama. Sementara itu, kultivar Barhe menghasilkan 5 bibit teridentifikasi sebagai jantan dan 5 bibit sebagai betina. Terdapat ketidak-sesuaian antara karakterisasi fenotip dan genotip dalam mendeterminasi jenis kelamin yaitu pada 2 bibit (B12, B15) kultivar Barhe. Namun, berdasarkan uji Chi-square dan Cramer's V menunjukan bahwa analisis p-value pada kultivar KL 1 (0,012) dan Barhe (0,048) masih dapat diterima melalui adanya hubungan kuat dan asosiasi yang signifikan antara karakter fenotip dan molekuler.

Dates (Phoenix dactylifera) are plants classified under the Arecaceae family. Dates typically reach reproductive maturity between 4-5 years, so at the early seedling stage, it is not possible to determine the sex of male or female. The purpose of this study is to determine the sex of Dates through phenotypic characteristics and molecular analysis of the SRY gene validated using ISSR markers. A total of 10 seedlings from each cultivar, KL.1 and Barhe, were used as samples. This research method includes the propagation of Dates In Vivo, allowing phenotypic characterization through the structure of the radicle (coiled or uncoiled) and the structure of the leaf primordia (curved or uncurved). The determination of male and female was also performed molecularly using the molecular marker of the SRY gene (SRY-date F-R) and ISSR markers (IS_A02 and IS_A71). The phenotypic characterization results of Date cultivar KL. 1 produced 7 male seedlings (K1, K3, K4, K5, K12, K14, K15) and 3 females (K2, K6, K9). Meanwhile, the Barhe cultivar indicated 7 males which are seedlings B1, B2, B5, B8, B11, B12, B15 and 3 females which are B4, B6, B10. Additionally, molecular determination was performed where the results for cultivar KL 1 validated the phenotypic characterization with indications of the same seedling sex. Meanwhile, the Barhe cultivar produced 5 seedlings identified as male and 5 as female. There was a discrepancy between phenotypic and genotypic characterization in determining sex for 2 seedlings (B12, B15) of the Barhe cultivar. However, based on the Chi-square test and Cramer's V, the p-value analysis for cultivar KL 1 (0.012) and Barhe (0.048) is still acceptable through the strong relationship and significant association between phenotypic and molecular characteristics.
