Abstract

Peningkatan suhu perairan akibat perubahan iklim dapat memengaruhi keseimbangan fisiologis dan sistem imun organisme akuatik, termasuk kepiting bakau (Scylla serrata), terutama selama fase molting. Spesies ini memiliki sistem imun bawaan yang ditunjukkan melalui aktivitas hemosit dalam hemolimfa. Penelitian ini bertujuan untuk menganalisis pengaruh suhu lingkungan terhadap Total Hemocyte Count (THC) dan Differential Hemocyte Count (DHC) pada kepiting bakau fase molting dan nonmolting. Metode penelitian meliputi pengaturan suhu air ke dalam tiga perlakuan (22 °C, 28 °C, dan 34 °C), pengambilan hemolimfa untuk dianalisis jumlah THC dan DHC. DHC mencakup tiga jenis sel hemosit, yaitu sel hyalin, semigranular, dan granular. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktorial dan analisis statistic menggunakan uji Kruskal-Wallis untuk data THC dan Two-Way ANOVA untuk pengaruh suhu dan fase molting terhadap DHC. Hasil penelitian menunjukkan bahwa suhu memengaruhi jumlah THC secara signifikan (p < 0,05), dengan nilai tertinggi konsisten ditemukan pada suhu 28 °C. Sedangkan, suhu tidak berpengaruh signifikan terhadap rasio DHC (p > 0,05), tetapi fase molting berpengaruh signifikan terhadap komposisi jenis sel hemosit (p < 0,05), dengan dominasi sel hyalin pada fase intermolt dan dominasi sel granular dan semigranular pada fase premolt. Oleh karena itu, dapat disimpulkan bahwa suhu lingkungan memengaruhi respons imun bawaan kepiting bakau melalui perubahan jumlah THC, sedangkan fase molting lebih berperan dalam mengatur komposisi DHC. Perubahan profil hemosit pada S. serrata menggambarkan adaptasi sistem imun terhadap perubahan suhu dan fase fisiologis dalam siklus hidupnya.

Increased water temperatures due to climate change can affect the physiological balance and immune systems of aquatic organisms, including mangrove crabs (Scylla serrata), especially during the molting phase. This species has an innate immune system that is demonstrated through the activity of hemocytes in the hemolymph. This study aims to analyze the effect of environmental temperature on Total Hemocyte Count (THC) and Differential Hemocyte Count (DHC) in molting and nonmolting mud crabs. The research methods included setting water temperature into three treatments (220C, 280C, and 340C), collecting hemolymph for THC and DHC analysis. DHC includes three types of hemocyte cells, hyaline, semigranular, and granular cells. This study employed a completely randomized design (CRD) factorial and statistical analysis using the Kruskal-Wallis test for THC data and Two-Way ANOVA for the effects of temperature and molting phase on DHC. The results showed that temperature significantly affected THC levels (p < 0.05), with the highest values consistently found at 28°C. However, temperature did not significantly affect the DHC ratio (p > 0.05), but molting phase significantly affected the composition of hemocyte cell types (p < 0.05), with hyaline cells dominating during the intermolt phase and granular and semigranular cells dominating during the premolt phase. Therefore, it can be concluded that environmental temperature affects the innate immune response of mangrove crabs through changes in THC levels, while the molting phase plays a greater role in regulating DHC composition. Changes in the hemocyte profile of S. serrata illustrate the adaptation of the immune system to changes in temperature and physiological phases in its life cycle.
