Abstract

Teripang merupakan salah satu hewan laut yang termasuk Echinodermata dengan kandungan protein yang cukup tinggi, yakni sebesar 40%. Tingginya kandungan protein pada teripang berpotensi dimanfaatkan sebagai sumber kolagen. Penelitian ini bertujuan untuk mengevaluasi pengaruh aplikasi kolagen teripang bola (Phyllophorus sp.) yang diekstraksi menggunakan enzim pepsin terhadap penyembuhan luka kulit mencit (Mus musculus) berdasarkan pengamatan morfologis dan histologis secara kualitatif, kuantitatif, dan semi-kuantitatif. Metode penelitian meliputi aklimatisasi mencit jantan strain DDY selama satu minggu, pemberian anestesi intramuskular menggunakan zoletil 50 mg/mL, dan pembuatan luka eksisi berbentuk lingkaran (diameter 4 mm) pada punggung mencit. Perlakuan dilakukan dengan mengaplikasikan kolagen teripang bola (Phyllophorus sp.) hasil ekstraksi pepsin sebanyak 2 mg pada luka secara in vivo selama 21 hari dengan variasi frekuensi pemberian 3 hari sekali dan 7 hari sekali. Pengamatan morfologi luka dilakukan setiap 2 hari sekali menggunakan aplikasi Image Raster 3, sementara jaringan luka diambil untuk dianalisis histologis setelah hari ke-21. Preparat histologi dibuat menggunakan metode pewarnaan Masson’s Trichrome dan diamati dengan mikroskop yang terhubung optilab. Hasil penelitian pemberian kolagen teripang bola (Phyllophorus sp.) hasil ekstraksi pepsin secara kualitatif berupa makroskopis morfologi dan histologis jaringan yang ditunjukkan melalui percepatan proses penutupan luka, pembentukan keropeng, serta pertumbuhan rambut pada area luka di hari ke-22. Sedangkan secara kuantitatif, berupa pertambahan luas penutupan luka dan persentase serat kolagen, dimana jumlah persentase serat kolagen H7 lebih tinggi dibandingkan kelompok H3 dan K0 yaitu sekitar (33,68±2,52%) sedangkan H3 = (29,25±3,02%) dan K0 = (25,44±0,76%). Serta hasil analisis semi-kuantitatif yaitu, rata-rata skor profil serat kolagen pada perlakuan H7 lebih tinggi dibandingkan kelompok perlakuan H3 dan K0 yaitu skor 13. Sementara skor profil serat kolagen pada H3 = 12,3 dan K0 = 9,7. Dari penelitian ini, hasil pemberian kolagen teripang bola (Phyllophorus sp.) dengan interval setiap 7 hari sekali (H7) dinilai lebih efektif dalam proses penyembuhan luka kulit mencit.

Sea cucumber is a marine animal belonging to Echinodermata with a fairly high protein content, namely 40%. The high protein content in sea cucumbers has the potential to be used as a source of collagen. This study aims to evaluate the effect of the application of sea cucumber (Phyllophorus sp.) collagen extracted using pepsin enzymes on the healing of skin wounds in mice (Mus musculus) based on qualitative, quantitative, and semi-quantitative morphological and histological observations. The research method includes acclimatization of male mice strain DDY for one week, administration of intramuscular anesthesia using 50 mg/mL zoletil, and creation of a circular excision wound (4 mm in diameter) on the back of the mice. The treatment was carried out by applying 2 mg of pepsin-extracted sea cucumber (Phyllophorus sp.) collagen to the wound in vivo for 21 days with varying frequency of administration once every 3 days and once every 7 days. Wound morphology observation was carried out every 2 days using the Image Raster 3 application, while the wound tissue was taken for histological analysis after day 21. Histological preparations were made using the Masson's Trichrome staining method and observed with a microscope connected to optilab. The results of the study of the administration of collagen from ball sea cucumbers (Phyllophorus sp.) extracted from pepsin qualitatively were in the form of macroscopic morphology and histology of the tissue shown by the acceleration of the wound closure process, scab formation, and hair growth in the wound area on day 22. While quantitatively, in the form of an increase in the area of wound closure and the percentage of collagen fibers, where the percentage of H7 collagen fibers was higher than the H3 and K0 groups, namely (33.68 ± 2.52%) while H3 = (29.25 ± 3.02%) and K0 = (25.44 ± 0.76%). As well as the results of semi-quantitative analysis, namely, the average score of collagen fiber profile in the H7 treatment was higher than the H3 and K0 treatment groups, namely a score of 13. While the collagen fiber profile score in H3 = 12.3 and K0 = 9.7. From this study, the results of administering sea cucumber collagen (Phyllophorus sp.) with an interval of once every 7 days (H7) were considered more effective in the healing process of mouse skin wounds
