Abstract
Luka merupakan terputusnya kontinuitas jaringan kulit, yaitu pada lapisan epitel kulit, serta lapisan paling dalam seperti jaringan subkutis, otot, maupun lemak. Kondisi ini terjadi akibat trauma atau tekanan eksternal. Luka harus diberi obat agar proses penyembuhan luka lebih cepat dan terhindar dari infeksi. Namun, metode pengobatan berasal dari biomaterial masih terbatas. Oleh karena itu, penelitian ini bertujuan untuk mengetahui pengaruh hasil ekstraksi asam asetat dari kolagen teripang bola (Phyllophorus sp.) terhadap proses kesembuhan luka kulit mencit (Mus musculus), dengan melalui pengamatan morfologi dan histologis secara kualitatif, kuantitatif, dan semi-kuantitatif. Penelitian dilakukan pada mencit strain DDY yang diaklimatisasi selama satu minggu. Luka eksisi berbentuk lingkaran (4 mm) dibuat menggunakan skin biopsy punch, kemudian diaplikasikan kolagen teripang hasil ekstraksi asam asetat sebanyak 2mg selama 21 hari dengan frekuensi pemberian tiga hari sekali (H3), tujuh hari sekali (H7), dan tanpa perlakuan (K0). Morfologi luka diamati menggunakan aplikasi Image Raster3, sementara jaringan luka diwarnai dengan Masson’s Trichrome untuk analisis histologis. Secara kualitatif, kelompok H3 dan H7 menunjukkan percepatan penutupan luka, pembentukan keropeng, dan munculnya folikel rambut pada hari ke-22. Secara kuantitatif, persentase serat kolagen tertinggi pada pada H3, dengan nilai masing-masing yaitu (30,66 ± 1,46%), H7 (29,75 ± 1,19%), dan K0 (25,44 ± 0,76%). Pada analisis semi-kuantitatif memperlihatkan peningkatan skor kolagen tertinggi pada H3 dengan nilai H3 (13) dan H7 (12,7), dan K0 (9,7). ========================================================================================================================
A wound is a break in the continuity of skin tissue, namely in the epithelial layer of the skin, as well as the deepest layers such as subcutaneous tissue, muscle, and fat. This condition occurs due to trauma or external pressure. Wounds must be treated with medication to accelerate the wound healing process and avoid infection. However, treatment methods derived from biomaterials are still limited. Therefore, this study aims to determine the effect of acetic acid extraction from sea cucumber (Phyllophorus sp.) collagen on the healing process of mouse (Mus musculus) skin wounds, through qualitative, quantitative, and semi-quantitative morphological and histological observations. The study was conducted on DDY strain mice that were acclimatized for one week. A circular excision wound (4 mm) was made using a skin biopsy punch, then 2 mg of acetic acid-extracted sea cucumber collagen was applied for 21 days with a frequency of administration every three days (H3), every seven days (H7), and without treatment (K0). Wound morphology was observed using the Image Raster3 application, while the wound tissue was stained with Masson's Trichrome for histological analysis. Qualitatively, H3 and H7 groups showed accelerated wound closure, scab formation, and hair follicle emergence on day 22. Quantitatively, the highest percentage of collagen fibers was at H3, with values of (30.66±1.46%), H7 (29.75±1.19%), and K0 (25.44±0.76%), respectively. Semi-quantitative analysis showed the highest increase in collagen scores at H3 with values of H3 (13) and H7 (12.7), and K0 (9.7).
