Abstract
Sludge dari Palm Oil Mill Effluent (POME) merupakan limbah pengolahan kelapa sawit yang mengandung senyawa organik tinggi dan berpotensi dimanfaatkan sebagai sumber nutrisi bagi mikroorganisme, khususnya bakteri selulolitik. Bakteri ini mampu mendegradasi selulosa menjadi gula sederhana yang dapat dikonversi menjadi gas hidrogen melalui fermentasi gelap. Penelitian ini bertujuan untuk mengkarakterisasi bakteri selulolitik penghasil biohidrogen dari sludge POME dengan pendekatan taksonomi polifasik, meliputi uji fenotipik dan analisis sekuen gen 16S rRNA. Isolasi bakteri dilakukan dari sludge POME, diikuti dengan uji aktivitas selulolitik menggunakan Congo Red 0,1%, NaCl 1 M, dan uji DNS. Aktivitas produksi hidrogen dianalisis menggunakan Gas Chromatography-Thermal Conductivity Detector (GC-TCD). Hasil menunjukkan bahwa dua isolat, yaitu SP4 dan SP8, merupakan bakteri fakultatif anaerobik yang memiliki aktivitas selulolitik tertinggi pada jam ke-96, dengan aktivitas selulolitik masing-masing sebesar 12,141 μmol/menit/mL dan 31,213 μmol/menit/mL pada substrat POME, serta 5,454 μmol/menit/mL dan 12,141 μmol/menit/mL pada substrat CMC. Dalam uji produksi hidrogen, SP4 dan SP8 secara single culture mampu menghasilkan hidrogen dari glukosa sebesar 0,0662% dan 0,0049%. Produksi hidrogen meningkat signifikan dalam perlakuan co-culture, yaitu 10,6592% dan 12,0934% pada glukosa serta 0,168% dan 0,0025% pada CMC. Identifikasi molekuler melalui analisis filogenetik dan BLAST menunjukkan bahwa kedua isolat tergolong dalam spesies Bacillus cereus.

Sludge from Palm Oil Mill Effluent (POME) is a by-product of palm oil processing that contains high levels of organic compounds and has the potential to be utilized as a nutrient source for microorganisms, particularly cellulolytic bacteria. These bacteria are capable of degrading cellulose into simple sugars that can be converted into hydrogen gas through dark fermentation. This study aims to characterize biohydrogen-producing cellulolytic bacteria isolated from POME sludge using a polyphasic taxonomic approach, including phenotypic tests and 16S rRNA gene sequence analysis. Bacterial isolation was carried out from POME sludge, followed by cellulolytic activity tests using 0.1% Congo Red, 1 M NaCl, and the DNS assay. Hydrogen production activity was analyzed using Gas Chromatography-Thermal Conductivity Detector (GC-TCD). The results showed that two isolates, SP4 and SP8, are facultative anaerobic bacteria with the highest cellulolytic activity on 96 hours, with activities of 12.141 μmol/min/mL and 31.213 μmol/min/mL on POME substrate, and 5.454 μmol/min/mL and 12.141 μmol/min/mL on CMC substrate, respectively. In the hydrogen production test, SP4 and SP8 as single cultures were able to produce hydrogen from glucose at 0.0662% and 0.0049%, respectively. Hydrogen production significantly increased under co-culture treatment, reaching 10.6592% and 12.0934% on glucose and 0.168% and 0.0025% on CMC, respectively. Molecular identification through phylogenetic analysis and BLAST indicated that both isolates belong to the species Bacillus cereus.
