Abstract

Triptamin merupakan salah satu amina biogenik yang terbentuk secara alami melalui dekarboksilasi asam amino triptofan. Keberadaannya dalam produk pangan fermentasi, seperti kecap asin, dapat menjadi indikator kualitas dan keamanan pangan karena akumulasi triptamin yang berlebih bersifat toksik. Penelitian ini bertujuan mengembangkan metode deteksi triptamin yang cepat, murah, dan selektif menggunakan nanokristal organik berbasis senyawa fluoresen yaitu BHB (3-(2-benzotiazolil)-4-hidroksibenzena sulfonat) yang terimobilisasi pada Microfluidic Paper-Based Analytical Device (μPAD). Nanokristal BHB dipreparasi melalui metode represipitasi dan dikarakterisasi menggunakan Particle Size Analyzer (PSA) untuk mengidentifikasi ukuran partikel berada dalam skala nano. Material ini kemudian diimobilisasi pada area uji μPAD dan dianalisis terhadap triptamin dalam berbagai konsentrasi untuk menguji sensitivitas deteksi. Pengujian juga dilakukan terhadap berbagai amina biogenik untuk menguji selektivitasnya. Demonstrasi dilakukan untuk menguji kinerja sistem deteksi pada sampel nyata berupa kecap asin. Deteksi dilakukan berdasarkan perubahan warna fluoresensi yang diamati menggunakan sinar UV dan dianalisis secara kuantitatif dengan perangkat lunak ImageJ menggunakan model warna CIELAB. Hasil uji sensitivitas menunjukkan bahwa sistem ini mampu mendeteksi triptamin dengan perubahan warna fluoresensi yang jelas dan memberikan nilai batas deteksi (LOD) yang rendah berdasarkan perhitungan regresi linier. Hasil uji selektivitas menunjukkan BHB selektif terhadap triptamin, ditunjukkan dengan perubahan warna signifikan yang tidak diamati pada amina biogenik lain. Aplikasi pada sampel nyata menghasilkan perubahan warna yang mengindikasikan keberadaan triptamin, sehingga efektif dalam mendeteksi analit target pada matriks kompleks. Perangkat μPAD yang dimodifikasi dengan nanokristal BHB menunjukkan potensi kuat sebagai platform analitik sederhana, murah, dan portabel untuk deteksi triptamin.

Tryptamine is one of the biogenic amines that is formed naturally through the decarboxylation of the amino acid tryptophan. Its presence in fermented food products, such as soy sauce, can be an indicator of food quality and safety because excessive accumulation of tryptamine is toxic. This study aims to develop a rapid, low-cost, and selective tryptamine detection method using organic nanocrystals based on fluorescent compounds, namely BHB (3-(2 benzothiazolyl)-4-hydroxybenzene sulfonate) immobilized on a Microfluidic Paper-Based Analytical Device (μPAD). BHB nanocrystals were prepared through the reprecipitation method and characterized using a Particle Size Analyzer (PSA) to identify particle sizes on the nanoscale. This material was then immobilized in the μPAD test area and analyzed against tryptamine in various concentrations to test the detection sensitivity. Tests were also carried out on various biogenic amines to test their selectivity. A demonstration was carried out to test the performance of the detection system on real samples in the form of soy sauce. Detection was carried out based on the fluorescence color change observed using UV light and quantitatively analyzed with ImageJ software using the CIELAB color model. The sensitivity test results showed that this system was able to detect tryptamine with a clear fluorescence color change and provided a low limit of detection (LOD) value based on linear regression calculations. The selectivity test results showed that BHB was selective for tryptamine, indicated by a significant color change that was not observed in other biogenic amines. Application to real samples resulted in a color change indicating the presence of tryptamine, making it effective in detecting target analytes in complex matrices. The μPAD device modified with BHB nanocrystals showed strong potential as a simple, inexpensive, and portable analytical platform for tryptamine detection.
