Abstract

Calkon dan pirazolina merupakan dua kelas senyawa yang dilaporkan memiliki aktivitas antikanker, dan sintesis pirazolina dapat menggunakan calkon sebagai prekursor. Penelitian yang dilakukan berhasil mendapatkan calkon (10) dengan rendemen 61 % dari reaksi kondensasi aldol yang melibatkan 3-asetilkumarin (3) dan furfural (5) pada kondisi basa. Reaksi calkon (10) dengan fenilhidrazina diperoleh pirazolina baru berupa senyawa 3-(5-(furan-2-il)-1-fenil-4,5-dihidro-1H-pirazol-3-il)-2H-kromen-2-on (9) dengan rendemen 20%. Uji kemurnian calkon (10) dan pirazolina (9) dilakukan dengan kromatografi lapis tipis dan titik leleh. Identifikasi struktur calkon (10) berdasarkan spektrum 1H NMR, sedangkan pirazolina (9) berdasarkan spektra ¹H-NMR, ¹³C-NMR, FTIR, dan massa resolusi tinggi.

Chalcones and pyrazolines are two classes of compounds reported to possess anticancer activity, and pyrazoline synthesis can utilize chalcones as precursors. This study successfully obtained chalcone (10) with a yield of 61% through an aldol condensation reaction involving 3-acetylcoumarin (3) and furfural (5) under basic conditions. The reaction of chalcone (10) with phenylhydrazine yielded a new chalcone-derived compound, 3-(5-(furan-2-yl)-1-phenyl-4,5-dihydro-1H-pyrazol-3-yl)-2H-chromen-2-one (9), with a yield of 20%. The purity of chalcone (10) and pyrazoline (9) was analyzed using thin-layer chromatography and melting point determination. Structural identification of chalcone (10) was based on ¹H NMR spectra, while pyrazoline (9) was characterized using ¹H NMR, ¹³C NMR, FTIR, and high-resolution mass spectrometry.
