Abstract
Bojonegoro merupakan daerah yang menyuplai produksi minyak sebesar 20% di Indonesia. Adanya kebutuhan masyarakat yang semakin tinggi dalam menggunakan minyak bumi menyebabkan semakin besar potensi adanya pencemaran lingkungan. Salah satu cara yang dapat digunakan untuk membantu menangani pencemaran ialah bioremediasi. Bioremediasi yang terjadi pada zona akar disebut rhizoremediasi. Pada rhizoremediasi terjadi asosiasi antara rhizosfer dengan mikroorganisme tanah melalui eksudasi senyawa organik sehingga terjadi peningkatan aktifitas mikroba pendegradasi minyak bumi. Oleh karena itu, tujuan dari penelitian ini ialah mengetahui profil mikrobioma rhizosfer rumput gajah (Pennisetum purpureum) yang tumbuh di tanah tercemar dan kurang tercemar, serta mengetahui pengaruh faktor fisikokimidan TPH (Total Petroleum Hydrocarbon) terhadap profil mikrobioma rhizosfer rumput gajah. Metode yang dilakukan dengan pengambilan data fisikokimia, sampel tanah tercemar dan rhizosfer dari rumput gajah. Kemudian dilakukan ekstraksi DNA dan pengujian metode gravimetri, NGS (Next Generation Sequencing) dan GCMS (Gas Chromatography Mass Spectrometry). Data dianalisis dengan IBM SPSS Statistic 22 untuk menguji normalitas dan korelasi data mikrobioma dengan data fisikokimia dan TPH (Total Petroleum Hydrocarbon) tanah. Selain itu, dilakukan analisis data indeks shannon wiener dan simpson yang akan dikorelasikan dengan data TPH (Total Petroleum Hydrocarbon) tanah. Hasil penelitian menunjukkan bahwa profil mikrobioma rhizosfer rumput gajah yang tumbuh di tanah tercemar dan kurang tercemar tidak menunjukkan perbedaan yang signifikan. Parameter fisikokimia dan TPH (Total Petroleum Hydrocarbon) mempengaruhi sebagian besar kelimpahan profil mikrobioma rhizosfer rumput gajah maupun indeks shannon wiener dengan pH yang paling mempengaruhi profil mikrobioma rhizosfer rumput gajah.

Bojonegoro is an area that supplies 20% of Indonesia's oil production. The existence of a higher public need in using petroleum causes the greater the potential for environmental pollution. One way that can be used to help deal with pollution is bioremediation. Bioremediation that occurs in the root zone is called rhizoremediation. In rhizoremediation there is an association between the rhizosphere and soil microorganisms through the exudation of organic compounds so that there is an increase in microbial activity that degrades petroleum. Therefore, the purpose of this study was to determine the rhizosphere microbiome profile of elephant grass (Pennisetum purpureum) growing on polluted and less polluted soil, and to determine the effect of physicochemical factors and TPH (Total Petroleum Hydrocarbon) on the rhizosphere microbiome profile of elephant grass. The method used is physicochemical data collection, contaminated soil samples and rhizosphere from elephant grass. Then DNA extraction and testing were carried out using gravimetric methods, NGS (Next Generation Sequencing) and GCMS (Gas Chromatography Mass Spectrometry). Data were analyzed with IBM SPSS Statistic 22 to test the normality and correlation of microbiome data with physicochemical data and soil TPH (Total Petroleum Hydrocarbon). In addition, an analysis of the Shannon Wiener and Simpson which would be correlated with the soil TPH (Total Petroleum Hydrocarbon) data. The results showed that the rhizosphere microbiome profile of elephant grass growing on polluted and less polluted soil did not show a significant difference. Physicochemical parameters and TPH (Total Petroleum Hydrocarbon) mostly affected the abundance of the elephant grass rhizosphere microbiome profile and the Shannon Wiener index with pH the most affecting the elephant grass rhizosphere microbiome profile.
