Abstract
Meningkatnya eksploitasi minyak bumi megakibatkan tingginya pencemaran tanah akibat tumpahan minyak bumi. Salah satu upaya rehabilitasi tanah tercemar minyak bumi yaitu menggunakan bakteri indigenous. Bakteri indigenous memiliki gen pendegradasi hidrokarbon sehingga dapat mendegradasi senyawa hidrokarbon yang merupakan senyawa utama dari minyak bumi. Penelitian ini bertujuan untuk mengetahui perbedaan kelimpahan gen pendegradasi hidrokarbon pada tanah yang memiliki tingkat pencemaran berbeda serta untuk mengetahui korelasi antara kelimpahan relatif gen pendegradasi hidrokarbon dengan sifat fisikokimia tanah di pertambangan minyak bumi tradisional Wonocolo, Kabupaten Bojonegoro Jawa Timur. Metode yang digunakan dalam penelitian ini yaitu pengambilan sampel tanah, ekstraksi DNA optimasi PCR, amplifikasi dengan qPCR serta analisis TPH menggunakan GCMS dan gravimetri. Hasil yang didapatkan dianalisa menggunakan uji statistik Pearson correlation dan ANOVA yang dilanjutkan dengan uji Tukey. Dari hasil penelitian yang didapatkan kelimpahan relatif gen paling tinggi pada tanah 1 yaitu gen assA. pada tanah 2.1 kelimpahan relatif gen yang banyak ditemukan yaitu alkB dan nahAc. Sedangkan gen C230 relatif tinggi pada setiap sampel tanah yang diteliti. Berdasarkan Uji korelasi, didapatkan bahwa gen alkB dan assA berkorelasi positif dengan kadar TPH dan didapatkan bahwa gen C230 berkorelasi negatif dengan kadar TPH. Sementara kelimpahan relatif gen nahAc, nahF dan PAH-RHD tidak menunjukan adanya korelasi dengan kadar TPH tanah. Kelimpahan relatif gen C230 dan nahAc juga berkorelasi negatif dengan salinitas tanah. Selain itu gen C230 juga menunjukan korelasi negatif dengan kadar air tanah. Korelasi positif ditunjukan oleh kelimpahan relatif gen assA terhadap kadar air tanah.

The increasing exploitation of petroleum has resulted in high soil pollution due to oil spills. One of the efforts to rehabilitate oil-contaminated soil is using indigenous bacteria. Indigenous bacteria have hydrocarbon degrading genes so that they can degrade hydrocarbon compounds which are the main compounds from petroleum. This study aims to determine differences in the abundance of microbiome genes in contaminated and unpolluted soils and to determine the correlation between the relatif abundance of hydrocarbon-degrading genes and oil physicochemical properties in the traditional oil mines of Wonocolo, Bojonegoro Regency, East Java. The methods used in this study were soil sampling, PCR optimization DNA extraction, amplification by qPCR and TPH analysis using GC-MS and gravimetry. The results obtained were analyzed using the Pearson correlation statistical test and the Tukey test. From the results of the study, it was found that the relatively highest gene in soil 1 was the assA gene. in soil 2.1 the most common relative genes found were alkB and nahAc. Meanwhile, the C230 gene was relatively high in each soil sample studied. Based on the correlation test, it was found that the alkB and assA genes were positively correlated with TPH levels and it was found that the C230 gene was negatively correlated with TPH levels. While the relative genes nahAc, nahF and PAH-RHD did not show any correlation with soil TPH levels. The relative abundance of C230 and nahAc genes was also negatively correlated with soil salinity. In addition, the C230 gene also showed a negative correlation with soil water content. The positive correlation shown by the relative relative of the assA gene in soil moisture content
