Abstract
Salah satu hambatan dalam budidaya tanaman Marasi (Curculigo latifolia) ini adalah benih bersifat rekalsitran serta ketidakseragaman karakter antar aksesi tanaman. Upaya yang paling tepat dalam penyediaan bibit berkualitas secara cepat adalah menggunakan teknologi kultur jaringan. Teknik kultur jaringan diartikan sebagai perbanyakan tanaman dari bagian meristematik secara aseptik dengan bantuan zat pengatur tumbuh dan faktor lingkungan seperti cahaya. Penelitian ini bertujuan untuk mengetahui perbedaan konsentrasi zat pengatur tumbuh dan jenis cahaya terhadap organogenesis tanaman marasi (Curculigo latifolia) secara in vitro. Bahan tanam yang digunakan berupa biji Curculigo latifolia yang dikultur dalam media MS (Murashige & Skoog) dengan penambahan kombinasi zat pengatur tumbuh IBA (1mgL-1) dan BAP (2 mgL-1,), BAP tunggal (2 mgL-1) dan pemberian jenis cahaya yang berbeda, cahaya merah, biru dan putih. Parameter yang diamati adalah persentase eksplan tumbuh tunas dan tinggi tunas. Hasil penelitian menunjukan persentase eksplan biji C. Latifolia muncul tunas terbaik pada perlakuan medium MS0 (K) dan pada semua perlakuan cahaya dengan nilai persentase sebesar 100%. Pada perlakuan medium IBA + BAP (1) persentase eksplan biji C. Latifolia muncul tunas tertinggi pada perlakuan cahaya (U), dengan nilai sebesar 67%. Sedangkan pada perlakuan medium BAP 2 (2) persentase pertumbuhan hanya terjadi pada perlakuan cahaya (P) dan (U). Perlakuan cahaya merah tidak memberikan respon pertumbuhan tunas. Sementara itu, tinggi tunas planlet C. Latifolia terbaik pada perlakuan medium MS0 (K) dan pada semua perlakuan cahaya dengan rata-rata tinggi 1,5 cm – 1,87 cm. Sedangkan tinggi tunas planlet C. Latifolia paling rendah pada perlakuan medium BAP 2 (B) dengan perlakuan cahaya (M)

One of the obstacles in the cultivation of this Marasi (Curculigo latifolia) plant is the recalcitrant nature of the seeds and the non-uniformity of characters between plant accessions. The most appropriate effort in providing quality seeds quickly is using tissue culture technology. Tissue culture technique is defined as aseptic propagation of plants from meristematic parts with the help of growth regulators and environmental factors such as light. This study aims to determine differences in the concentration of growth regulators and types of light on the organogenesis of marasi plants (Curculigo latifolia) in vitro. Planting material used in the form of seeds of Curculigo latifolia cultured in MS media (Murashige & Skoog) with the addition of a combination of growth regulators IBA (1 mgL-1) and BAP (2 mgL-1,), single BAP (2 mgL-1) and giving different types of light, red, blue and white light. The parameters observed were the percentage of explants growing shoots and shoot height. The results showed that the percentage of C. Latifolia seed explants showed the best shoots in the MS0 (K) medium treatment and in all light treatments with a percentage value of 100%. In the medium treatment IBA + BAP (1) the percentage of seed explants of C. Latifolia showed the highest shoots in the light treatment (U), with a value of 67%. While in the treatment of BAP 2 medium (2) the percentage growth only occurred in the light treatment (P) and (U). Red light treatment did not respond to shoot growth. Meanwhile, the shoot height of C. latifolia plantlets was best in the MS0 (K) medium treatment and in all light treatments with an average height of 1.5 cm – 1.87 cm. While the plantlet shoot height of C. Latifolia was the lowest in the treatment of BAP 2 medium (B) with light treatment (M)
