Abstract
Teknologi enzimatik, bagian ilmu biologi dengan kajian produksi dan aplikasi enzim di berbagai bidang industri dalam rangka meningkatkan keberlangsungan usaha dalam suatu industri, termasuk industri farmasi. Salah satu peluang penerapan teknologi enzimatik di sektor ini adalah modifikasi polimer penghantar obat berbasis administrasi oral. Sistem penghantaran obat telah berkembang sejak tahun 1952, dengan pengembangan enkapsulasi untuk peningkatan durasi kerja obat. Perkembangan berikutnya berlanjut pada tahun 1990 dengan modifikasi permukaan menggunakan poli etilen glikol. Hingga saat ini, beragam variasi ukuran, material, modifikasi permukaan, dan bentuk dimanfaatkan untuk peningkatan kerja obat, termasuk modifikasi berbasis enzimatik. Dalam bidang ini, enzim digunakan untuk memodifikasi polimer yang berperan sebagai pengangkut obat atau drug transporter guna meningkatkan indeks terapeutik obat, yang merupakan efek yang ditimbulkan suatu obat sesuai dengan dosis pemberian, dan kecukupan dosis obat. Aplikasinya dapat dilihat pada sistem penghantaran obat 4-ASA untuk penanganan IBD dengan modifikasi polimer berbasis enzim transglutaminase secara in vitro, dan modifikasi polimer sensitif enzim berbasis azoreduktase dan esterase dengan peningkatan 97%. Studi literatur ini memberikan gambaran tentang peluang teknologi enzim dalam memodifikasi polimer penghantaran obat oral. Teknik modifikasi enzimatik untuk penghantaran obat mampu meningkatkan ketersediaan biologis dan kerja obat secara in vivo maupun in vitro. Modifikasi didasarkan pada kesesuaian jenis enzim dan polimer. Keunggulan yang didapat antara lain lain meningkatkan spesifitas obat, menurunkan efek toksisitas, dan mencegah pelepasan obat secara prematur.

Enzymatic technology is part of biological sciences with studies on the production and application of enzymes in various industrial fields to improve business sustainability in industry, including the pharmaceutical industry. One of the opportunities for the application of enzymatic technology in this sector is polymer modification of drugs based on oral administration. Drug delivery systems have evolved since 1952, with the development of encapsulations to increase drug duration. Subsequent developments continued in 1990 with surface modification using polyethylene glycol. Until now, various sizes, materials, surface modifications, and forms of use for drug enhancement, including enzymatic-based modifications have been found. In this art, enzymes are used to modify polymers that act as drug transporters or drug transporters to increase the therapeutic index of the drug, which is the effect that the drug produces according to the dose of administration, and the adequacy of the drug dose. Its application can be seen in the 4-ASA drug delivery system for the treatment of IBD with polymer modification based on the transglutaminase enzyme in vitro, and modification of the enzyme-sensitive polymer based on azoreductase and esterase with an increase of 97%. This literature study provides an overview of the opportunities of enzyme technology in modifying oral drug delivery polymers. Enzymatic modification techniques for drug delivery can increase the biological availability and action of drugs in vivo and in vitro. Modification is based on the type of enzyme and polymer. The advantages include increasing the specificity of the drug, reducing the effect of toxicity, and preventing prematur drug acquisition.
