Abstract

Mikroorganisme laut memiliki respon metabolisme dan fisiologis yang unik dikarenakan faktor lingkungan yang sangat bervariasi, salah satunya yaitu menghasilkan senyawa bioaktif. Salah satu bioaktif yang dihasilkan yaitu lipopeptida yang termasuk ke dalam biosurfaktan yang memiliki karakteristik antimikroba, ramah lingkungan, dan biodegradabilitas tinggi, serta memiliki stabilitas yang tinggi terhadap suhu, pH, dan salinitas yang ekstrim. Sehingga eksplorasi produksi lipopeptida terus dilakukan. Penelitian ini bertujuan untuk mengetahui potensial antibiotik ekstrak lipopeptida dari isolat LBF-1-0141 Alcanivorax xenomutans. A. xenomutans merupakan salah satu bakteri laut yang diketahui dapat mendegradasi hydrocarbon. Pembuktian potensi antibakteri dari metabolit lipopeptida A. xenomutans dilakukan dengan paper disk diffusion dan minimum inhibitory concentration dengan konsentrasi 10 mg/mL, 5 mg/mL, dan 2,5 mg/mL terhadap bakteri Staphylococcus aureus dan Escherichia coli. Ekstrak lipopeptida A. xenomutans memiliki aktivitas antibakteri terhadap S. aureus dengan diameter zona bening paling besar dihasilkan oleh konsentrasi 10 mg/mL yaitu 9,8±0,10 mm, diikuti 8,7±0,02 mm oleh konsentrasi 5 mg/mL, dan 6,63±0,04 mm oleh konsentrasi 2,5 mg/mL. Namun, tidak terbentuk zona bening ekstrak lipopeptida terhadap E. coli. Pertumbuhan S. aureus dengan penambahan ekstrak lipopeptida masing-masing konsentrasi 10 mg/mL, 5 mg/mL, 2,5 mg/mL yaitu 68%, 84%, dan 92% terhadap pertumbuhan bakteri tanpa perlakuan. Sedangkan pada E. coli yaitu 94%, 100%, dan 98% pada masing-masing konsentrasi. Sehingga dapat disimpulkan bahwa ekstrak lipopeptida A. xenomutans memiliki efek bakteriostatik hanya terhadap S. aureus dan semakin tinggi konsentrasi ekstrak lipopeptida A. xenomutans yaitu dari 2,5 mg/mL hingga 10 mg/mL, maka semakin besar daya hambat yang dihasilkan terhadap S. aureus.

Marine microorganisms have unique metabolic and physiological responses due to highly varied environmental factors, one of which is the production of bioactive compounds. One of the bioactive produced is lipopeptide, a biosurfactant that has antimicrobial characteristics, is environmentally friendly, has high biodegradability, and has high resistance to extreme temperatures, pH, and salinity. So that the exploration of lipopeptide production continues. This study aimed to determine the antibiotic potency of lipopeptide extract from isolate LBF-1-0141 Alcanivorax xenomutans. A. xenomutans is one of the marine bacteria known to degrade hydrocarbons. Proof of the antibacterial potential of lipopeptide metabolites of A. xenomutans was carried out by paper disk diffusion and the minimum inhibitory concentrations were 10 mg/mL, 5 mg/mL, and 2.5 mg/mL against Staphylococcus aureus and Escherichia coli bacteria. The lipopeptide extract of A. xenomutans had antibacterial activity against S. aureus with the most extensive zone diameter produced by a concentration of 10 mg/mL, which was 9.8±0.10 mm, followed by 8.7±0.02 mm by a concentration of 5 mg/mL, and 6.63±0.04 mm by a concentration of 2.5 mg/mL. However, there was no clear zone of lipopeptide extract against E. coli. The growth of S. aureus with the addition of lipopeptide extract with concentrations of 10 mg/mL, 5 mg/mL, and 2.5 mg/mL, respectively, were 68%, 84%, and 92% of the growth of bacteria without treatment. Meanwhile, for E. coli, they were 94%, 100%, and 98% at each concentration. So that it can be said that the lipopeptide extract of A. xenomutans has a bacteriostatic effect only against S. aureus and increases the concentration of the lipopeptide extract of A. xenomutans from 2,5 mg/mL to 10 mg/mL, the greater the inhibitory power produced against S. aureus.
