Abstract

Aktivitas industri perminyakan di area Tambang Minyak Bumi Wonocolo, Bojonegoro, Jawa Timur umumnya menghasilkan limbah crude oil yang dapat menyebabkan pencemaran lingkungan. Namun, terdapat beberapa tumbuhan yang mampu hidup dengan baik di area pertambangan tersebut, salah satunya adalah tanaman kersen (Muntingia calabura L.). Tanaman kersen (M. calabura L.) mampu hidup dengan baik di iklim tropis dan di berbagai cekaman kontaminan. Pada akar dari tanaman kersen (M. calabura L.) terdapat mikroorganisme tanah yang berasosiasi dengan rhizosfer. Mikroorganisme tersebut kemungkinan memiliki potensi dalam mendegradasi hidrokarbon. Akan tetapi, mikroorganisme tanah apa saja yang ada di daerah rhizosfer tersebut belum diketahui. Sehingga perlu penelitian lebih lanjut untuk menganalisis kelimpahan dan diversitas mikrobioma rhizosfer tanaman kersen (M. calabura L.). Metode penelitian ini meliputi pengambilan sampel tanah, sampel rhizosfer, dan data sifat fisikokimia tanah, ekstraksi DNA, uji kemurnian dan konsentrasi DNA, analisis metagenomik menggunakan Next Generation Sequencing (NGS), analisis kandungan TPH, dan analisis data. Analisis perbedaan profil mikrobioma pada titik tercemar dan kurang tercemar dilakukan menggunakan uji T pada software IBM SPSS Statistics for windows. Analisis korelasi profil mikrobioma dengan sifat fisikokimia tanah dan TPH menggunakan software R-studio. Uji statistik dilakukan pada taraf kepercayaan 95% (α = 0,05). Hasil dari perbedaan profil mikrobioma rhizosfer tanaman kersen (M. calabura L.) di tanah tercemar dan kurang tercemar menunjukkan bahwa kelimpahan relatif pada taksa filum, Filum Chloroflexi dan Proteobacteria terdapat perbedaan signifikan antara titik tercemar dan kurang tercemar. Pada taksa kelas, Kelas Gammaproteobacteria terdapat perbedaan signifikan antara titik tercemar dan kurang tercemar. Pada taksa ordo, famili, genus, dan spesies tidak terdapat perbedaan signifikan antara titik tercemar dan kurang tercemar. Indeks diversitas Shannon-Wiener dan Simpson tidak ditemukan adanya perbedaan signifikan antara titik tercemar dan kurang tercemar. Hasil analisis korelasi profil mikrobioma dengan sifat fisikokimia tanah dan TPH diketahui bahwa kelimpahan relatif Filum Proteobacteria dan Chloroflexi, Kelas Gammaproteobacteria, Acidobacteria_Gp6 dan Acidobacteria_Gp3, Ordo Xanthomonadales, Famili Sinobacteriaceae, Genus Acidobacter_Gp6 berkorelasi signifikan. Pada taksa spesies tidak ditemukan adanya kelimpahan relatif yang berkorelasi signifikan dengan sifat fisikokimia tanah dan TPH. Indeks diversitas Shannon-Wiener dan Simpson tidak berkorelasi signifikan.

Oil industry activities in the Wonocolo Petroleum Mine area, Bojonegoro, East Java generally produce crude oil waste, which can be causing of environmental pollution. But, there are several plants that are able to live well in the mining area, the one is cherry tree (Muntingia calabura L.). The cherry plant (M. calabura L.) is able to live well in tropical climates and in various contaminant stresses. At the roots of the cherry plant (M. calabura L.), there are soil micro-organisms associated with the rhizosphere. These micro-organisms may have potentially degrade of hydrocarbons. Whereas, the kind of soil micro-organisms that exist in the rhizosphere area is not yet known. Therefore, need continuous research to analyze the abundance and diversity of the cherry (M. calabura L.) rhizosphere microbiome. This research method includes soil sampling, rhizosphere samples, and soil physicochemical properties data; DNA extraction; DNA purity and concentration tests; metagenomic analysis using Next Generation Sequencing (NGS); TPH content analysis; and data analysis. Differential analysis of microbiome profiles at the polluted and less polluted was carried out using the T test on the IBM SPSS Statistics for Windows software. Correlation analysis of microbiome profiles with soil physicochemical properties and TPH using R-studio software. Statistical tests conducted at the 95% of confidence level (α = 0.05). The differences result in the rhizosphere microbiome profile of cherry (M. calabura L.) in polluted and less polluted soils showing the relative abundance of the taxa phylum, Phylum Chloroflexi, and Proteobacteria be found a significant difference between polluted and less polluted soils. In the taxa class, Gammaproteobacteria class there is a significant difference point between polluted and less polluted soils. In taxa order, family, genus, and species, there is no significant difference point between polluted and less polluted soils. The Shannon-Wiener and Simpson diversity indices discovered no statistically significant difference between polluted and less polluted soils. The result of correlation analysis of the microbiome profile with soil physicochemical properties and TPH showing the relative abundance of Phylum Proteobacteria and Chloroflexi, Gammaproteobacteria class, Acidobacteria_Gp6 and Acidobacteria_Gp3, Order Xanthomonadales, Family Sinobacteriaceae, Genus Acidobacter_Gp6 correlated significantly. There is no relative abundance found in the species taxa, which is significantly correlated with soil physicochemical properties and TPH. The Shannon-Wiener and Simpson diversity indexes are not significantly correlated.
