Abstract
Stevia rebaudiana merupakan sumber dari steviol glikosida dengan tingkat kemanisan 250-300 kali lebih manis daripada gula tebu, sehingga stevia dapat dimanfaatkan sebagai pemanis alami. Budidaya kultur in vitro stevia yang efektif masih dalam pengembangan. Salah satunya dapat dilakukan dengan penambahan sukrosa dan perlakuan jenis cahaya. Tujuan dari penelitian ini adalah mengetahui pengaruh jenis cahaya dan konsentrasi sukrosa terhadap pertumbuhan kultur in vitro stevia. Bahan tanam yang digunakan berupa pucuk tunas dari Stevia rebaudiana yang dikultur dalam media pertumbuhan dengan konsentrasi sukrosa 0%, 3%, dan 5% pada cahaya putih, biru, merah, dan hijau. Parameter yang diamati adalah tinggi tanaman dan jumlah daun. Hasil penelitian menunjukkan pertumbuhan tinggi tanaman terbaik pada perlakuan cahaya hijau dan konsentrasi sukrosa 3% (HS2) dengan nilai sebesar 2.36 cm, namun daun berwarna pucat. Sedangkan pada parameter jumlah daun hasil terbaik diperoleh pada perlakuan cahaya merah dan sukrosa 3% (MS2) dengan nilai sebesar 10.83 helai. Sedangkan kombinasi perlakuan cahaya dan sukrosa untuk mendapatkan bibit stevia yang unggul pada tinggi tanaman dan jumlah daun ditunjukkan pada perlakuan kombinasi cahaya merah dan sukrosa 3% (MS2).

Stevia rebaudiana is a source of steviol glycosides with a sweetness level of 250-300 times sweeter than cane sugar, so stevia can be used as a natural sweetener. Effective in vitro culture of stevia is still under development. One of them can be done by adding sucrose and light treatment. The purpose of this study was to determine the effect of the type of light and the concentration of sucrose on the growth of stevia culture in vitro. The planting material used was shoots of Stevia rebaudiana cultured in growth media with 0%, 3%, and 5% sucrose concentrations in white, blue, red, and green light. Parameters observed were plant height and the number of leaves. The results showed the best plant height growth in green light treatment and 3% sucrose concentration (HS2) with a value of 2.36 cm, but the leaves were pale. While on the number of leaves the best results were obtained in the treatment of red light and 3% sucrose (MS2) with a value of 10.83 strands. Meanwhile, the combination of light and sucrose treatment to obtain superior stevia seedlings in plant height and number of leaves was shown in the combination treatment of red light and 3% sucrose (MS2).
