Abstract
Kebutuhan porang (A. muelleri Blume.) di Indonesia dan luar negeri setiap tahun terus meningkat. Peningkatan porang belum diimbangi budidaya tanaman secara luas sehingga ketersediaaan bibit masih rendah. Di sisi lain, perbanyakan tanaman porang memiliki kendala dengan masa dormansi yang panjang. Salah satu cara untuk menyediakan bibit secara cepat yaitu melalui kultur in vitro. Untuk meningkatkan pertumbuhan tanaman porang secara kultur in vitro dapat dilakukan dengan penambahan ZPT. ZPT (Zat Pengatur Tumbuh) yang dapat digunakan diantaranya sitokinin berupa BAP dan auksin berupa IAA. Kedua ZPT tersebut diharapkan mampu menstimulus terbentuknya tunas. Tujuan dari penelitian ini untuk mengetahui pengaruh kombinasi konsentrasi ZPT BAP dan IAA terhadap induksi tunas dari bulbil tanaman porang (A. muelleri Blume.) secara in vitro. Metode yang digunakan yaitu kultur jaringan dengan aplikasi ZPT. Analisis data dilakukan secara deskriptif kualitatif yang terdiri dari persentase pembentukan tunas, jumlah tunas, panjang tunas, dan hari terbentuk tunas. Hasil dari penelitian ini menunjukkan bahwa pada semua perlakuan konsentrasi BAP (6-Benzyl Amino Purine), IAA (Indole-3-Acetic Acid), serta kombinasi BAP dan IAA dapat menginduksi tunas dengan persentase sebesar 100%, pada perlakuan dengan aplikasi ZPT yang memberikan induksi tunas terbaik adalah pada perlakuan B1 (0,25 IAA) didapatkan rerata panjang tunas tertinggi yaitu 1,5 cm dengan rerata hari terbentuk tunas yaitu 4.75.

The need for porang (A. muelleri Blume.) in Indonesia and abroad continues to increase every year. An increase in the number of plants does not need to be planted widely so that the availability of seeds is still available. On the other hand, porang plant propagation has problems with a long dormancy period. One way to provide seeds quickly is through in vitro culture. To increase the growth of porang plants in vitro culture can be done with the addition of PGR. ZPT (Growth Regulating Substances) that can be used include cytokinins in the form of BAP and auxin in the form of IAA. The two ZPTs are expected to be able to stimulate the formation of tuna. The purpose of this study was to determine the effect of the combined concentration of BAP and IAA on the induction of tuna from bulbil porang (A. muelleri Blume.) in vitro. The method used is tissue culture with PGR application. Data analysis was carried out in a qualitative descriptive manner, consisting of the proportion of tuna formation, number of tuna, tuna length, and days of formation of tuna. The results of this study indicate that in all treatments the concentration of BAP (6-Benzyl Amino Purine), IAA (Indole-3-Acetic Acid), and the combination of BAP and IAA can induce tuna with a percentage of 100%, in the treatment with ZPT application which gives The best shoot induction was in treatment B1 (0.25 IAA) the highest average shoot length was 1.5 cm with the average day of shoot formation was 4.75.
