Abstract
Hiperinsulinemia merupakan keadaan kadar insulin yang tinggi dan menyebabkan terganggunya mekanisme pensinyalan insulin pada jalur translokasi glucose transporter type 4 (GLUT4). Gangguan pada pensinyalan tersebut mengakibatkan penurunan efisiensi penyerapan glukosa dan peningkatan kadar glukosa darah. Oleh karena itu, pemodelan matematis dan pengujian in-vitro diperlukan untuk memahami secara kuantitatif hubungan antar protein yang terlibat dalam jalur pensinyalan GLUT4. Penelitian ini bertujuan untuk memperoleh konsentrasi protein target insulin (INS) dan insulin receptor (INSR) serta interpretasi protein GLUT4 sebagai parameter kuantitatif. Sel otot sehat diberi perlakuan environment normal dan hiperinsulinemia serta lama waktu paparan 10,20,30,40, dan 50 menit. Pada hasil konsentrasi protein target INS dan INSR pada jalur pensinyalan insulin di sel otot sehat environment normal menunjukkan respons fisiologis yang normal dengan standar deviasi rendah. Pada environment hiperinsulinemia, protein INS memiliki nilai standar deviasi ± 4,766 mIU/L, sedangkan protein INSR memiliki nilai standar deviasi ± 4,673 mIU/L. Hasil tersebut menunjukkan adaptasi awal sel terhadap kondisi hiperinsulinemia sebagai respon awal resistensi insulin. Interpretasi pensinyalan GLUT4 pada jalur pensinyalan insulin di sel otot sehat kemungkinan terjadi translokasi karena nilai standar deviasi yang rendah. Namun, pada perlakuan hiperinsulinemia kemungkinan terjadi distranslokasi karena standar deviasi yang tinggi. Hal ini menyesuaikan hasil konsentrasi serangkaian protein target lainnya.

Hyperinsulinemia is a condition of high insulin levels and causes disruption of the insulin signaling mechanism in the glucose transporter type 4 (GLUT4) translocation pathway. This signaling disruption results in decreased glucose uptake efficiency and increased blood glucose levels. Therefore, mathematical modeling and in vitro testing are needed to quantitatively understand the relationship between proteins involved in the GLUT4 signaling pathway. This study aims to obtain the concentration of insulin target proteins (INS) and insulin receptor (INSR) and the interpretation of GLUT4 protein as a quantitative parameter. Healthy muscle cells were treated with normal and hyperinsulinemic environments with exposure times of 10, 20, 30, 40, and 50 minutes. The results of the concentration of INS and INSR target proteins in the insulin signaling pathway in healthy muscle cells in a normal environment showed a normal physiological response with a low standard deviation. In the hyperinsulinemic environment, the INS protein had a standard deviation value of ± 4.766 mIU/L, while the INSR protein had a standard deviation value of ± 4.673 mIU/L. These results indicate initial cell adaptation to hyperinsulinemia as the early response of insulin resistance. Interpretation of GLUT4 signaling in the insulin signaling pathway in healthy muscle cells suggests translocation is likely due to the low standard deviation. However, in hyperinsulinemia treatment, distranslocation is likely due to the high standard deviation. This may skew the concentration results of a series of other target proteins.
