Abstract

Tanaman bangun bangun (C. barbatus (Andrews) Benth. ex G.Don) merupakan salah satu tanaman herbal yang secara tradisional dimanfaatkan sebagai obat, namun kajian ilmiah mengenai aktivitas antioksidan dan profil metabolit sekundernya masih terbatas. Penelitian ini bertujuan untuk menganalisis aktivitas antioksidan serta mengidentifikasi profil metabolit sekunder yang terkandung dalam ekstrak metanol daun C. barbatus menggunakan metode Liquid Chromatography–Mass Spectrometry (LC-MS). Ekstraksi dilakukan dengan metode maserasi menggunakan 4 jenis pelarut untuk memperoleh ekstrak kasar. Aktivitas antioksidan diuji menggunakan metode peredaman radikal bebas, sedangkan analisis profil metabolit sekunder dilakukan menggunakan instrumen LC-MS untuk mengidentifikasi komponen kimia berdasarkan nilai mass-to-charge ratio (m/z) dan waktu retensi. Data hasil analisis kemudian dibandingkan dengan pustaka dan basis data senyawa untuk menentukan kemungkinan struktur senyawa yang terdeteksi. Hasil penelitian menunjukkan bahwa ekstrak metanol daun C. barbatus memiliki aktivitas antioksidan yang tergolong baik, yang mengindikasikan kemampuannya dalam menangkal radikal bebas. Analisis LC-MS berhasil mengidentifikasi berbagai metabolit sekunder yang didominasi oleh golongan asam organik, fenolat, flavonoid, dan senyawa bioaktif lainnya yang diketahui berkontribusi terhadap aktivitas antioksidan. Perbedaan profil senyawa dibandingkan penelitian sebelumnya diduga dipengaruhi oleh metode ekstraksi, kondisi analisis, serta perbedaan bagian tanaman yang digunakan. Berdasarkan hasil tersebut, dapat disimpulkan bahwa daun C. barbatus berpotensi sebagai sumber alami antioksidan dan memiliki kandungan metabolit sekunder yang beragam. Penelitian ini diharapkan dapat menjadi dasar ilmiah untuk pengembangan lebih lanjut tanaman bangun bangun sebagai bahan baku fitofarmaka atau produk kesehatan berbasis bahan alam.

Bangun bangun (C. barbatus (Andrews) Benth. ex G. Don) is a herbal plant traditionally used as a medicinal remedy. However, scientific studies on its antioxidant activity and secondary metabolite profile remain limited. This study aimed to evaluate the antioxidant activity and identify the secondary metabolite profile of methanolic extracts of C. barbatus leaves using Liquid Chromatography–Mass Spectrometry (LC–MS). Extraction was carried out by maceration using four different solvents to obtain crude extracts. Antioxidant activity was assessed using a free radical scavenging assay, while secondary metabolite profiling was performed using LC–MS to identify chemical components based on their mass-to-charge ratio (m/z) and retention time. The obtained data were compared with literature and compound databases to predict the possible structures of the detected compounds. The results showed that the methanolic extract of C. barbatus leaves exhibited good antioxidant activity, indicating its potential to scavenge free radicals. LC–MS analysis successfully identified various secondary metabolites, predominantly organic acids, phenolic compounds, flavonoids, and other bioactive compounds known to contribute to antioxidant activity. Differences in compound profiles compared to previous studies are presumed to be influenced by extraction methods, analytical conditions, and variations in the plant parts used. Based on these findings, C. barbatus leaves have potential as a natural source of antioxidants and possess a diverse secondary metabolite composition. This study is expected to provide a scientific basis for further development of bangun bangun as a raw material for phytopharmaceuticals or natural health-based products.
