Abstract

Penelitian ini dilakukan untuk mengetahui potensi ekstrak TKKS sebagai antibakteri terhadap bakteri Enterococcus faecalis dan Pseudomonas aeruginosa. Sebelum dilakukan pengujian, TKKS diekstraksi dengan metode maserasi selama 3 x 24 jam. Maserasi dilakukan menggunakan 4 pelarut yang berbeda, yaitu aquades, etanol 96%, aseton, dan etil asetat. Untuk memastikan adanya kandungan senyawa metabolit sekunder, pada penelitian ini dilakukan uji fitokimia terhadap senyawa yang bersifat sebagai antibakteri seperti flavonoid, saponin, tannin, alkaloid, steroid, dan terpenoid. Uji aktivitas antibakteri ditentukan secara kualitatif dengan metode difusi cakram dan secara kuantitatif dengan metode mikrodilusi. DMSO digunakan sebagai kontrol negatif dan kloramfenikol sebagai kontrol positif. Uji difusi cakram dilakukan dengan konsentrasi 125 mg/mL. Hasil uji difusi cakram pada bakteri E. faecalis tidak menunjukkan adanya zona hambat pada semua ekstrak TKKS, sedangkan pada bakteri P. aeruginosa menunjukkan adanya zona hambat pada ekstrak aseton konsentrasi 125 mg/mL dengan rata-rata 6 mm. Uji mikrodilusi dilakukan dengan 6 variasi konsentrasi ekstrak aseton dan kontrol positif, yaitu 500 mg/mL, 250 mg/mL, 125 mg/mL, 75 mg/mL, 50 mg/mL, dan 25 mg/mL dan kontrol negatif. Hasil uji mikrodilusi menunjukkan bahwa ekstrak aseton TKKS mampu menghambat pertumbuhan bakteri P. aeruginosa hingga 60,24% pada konsentrasi 500 mg/mL dengan konsentrasi IC50 sebesar 369,79 mg/mL. Konsentrasi hambat minimum (KHM) yang diperoleh sebesar 53,34 mg/mL.

This study aims to determine the potential of extracting empty fruit bunches as an antibacterial against Enterococcus faecalis and Pseudomonas aeruginosa bacteria. Before any evaluation was done, EFB were extracted by maceration for 3 x 24 hours. There were 4 different solvents used in maceration i.e. distilled water, ethanol 96%, acetone, and ethyl acetate. To ensure that secondary metabolites are present, phytochemical constituents were evaluated concerning compounds that have antibacterial properties such as flavonoid, saponin, tannin, alkaloid, steroid, and terpenoid. Antibacterial assays were determined by disk diffusion as a qualitative assay and micro dilution as a quantitative assay. DMSO was used as a negative control and chloramphenicol as a positive control. Concentration that was used is 125 mg/mL. E. faecalis show no inhibition for all EFB extract in disk diffusion. On the other hand, acetone extract showed inhibition on 125 mg/mL on an average 6 mm on P aeruginosa. There were 6 concentrations of acetone extract and positive control were evaluated in micro dilution assay i.e. 500 mg/mL, 250 mg/mL, 125 mg/mL, 75 mg/mL, 50 mg/mL, and 25 mg/mL and also negative control. Micro dilution assay showed that the acetone extract of EFB was capable of inhibiting the growth of P. aeruginosa bacterium by 60,24% on 500 mg/mL and the IC50 is 369,79 mg/mL. Minimum inhibition concentration (MIC) is 53,34 mg/mL.
