Abstract

Mikroalga Picochlorum sp. berpotensi tinggi sebagai bahan biodiesel dan penghasil karotenoid. Walne adalah pupuk sintetik yang sering digunakan dalam kultur mikroalga yang mengandung sumber nutrisi tinggi serta dilengkapi dengan kebutuhan makronutrien dan mikronutrien untuk pertumbuhan dan perkembangan mikroalga. Namun, harga pupuk walne yang mahal membuat kultivasi mikroalga membutuhkan biaya yang tinggi. Mikroalga dapat dikultivasi secara heterotrofik dengan menambahkan nutrisi yang berasal dari limbah organik untuk mendapatkan biomassa yang melimpah serta dapat menekan biaya produksi. Salah satu limbah organik yang berpotensi sebagai nutrisi tambahan dalam kultivasi heterotrofik adalah Fermented Mother Liquor (FML). FML merupakan limbah cair dari pembuatan bahan penyedap makanan yang memiliki kandungan crudeprotein dan nitrogen yang tinggi. Tujuan dari penelitian ini adalah untuk mengetahui berat kering biomassa, jumlah sel, kecepatan tumbuh, profil asam lemak, konsentrasi lipid dan karotenoid dari Picochlorum sp. dengan variasi komposisi medium FML dan walne, yaitu P1 (0% FML + walne), P2 (0,01% FML + walne), P3 (0,01% FML), P4 (0,03% FML + walne), P5 (0,03% FML), P6 (0,06% FML + walne), dan P7 (0,06% FML). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) satu faktor dengan 4 kali pengulangan. Parameter yang diamati adalah biomass, jumlah sel, konsentrasi lipid, dan konsentrasi karotenoid yang dianalisis secara kuantitatif, sedangkan profil asam lemak dianalisis secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa variasi komposisi medium FML dan walne memberikan pengaruh terhadap berat kering biomassa, kecepatan tumbuh, konsentrasi lipid dan karotenoid Picochlorum sp. Biomassa dan kecepatan tumbuh tertinggi diperoleh pada P6 (0,06% FML + walne) sebesar 0,115 gram dan 299 sel/hari. Konsentrasi lipid tertinggi diperoleh pada perlakuan P2 (0,01% FML + walne) sebesar 29, 62%. Sedangkan, konsentrasi karotenoid tertinggi diperoleh pada perlakuan P3 (0,01% FML) sebesar 36,104 μg/mL. Hasil profil asam lemak perlakuan P1 (0% FML + walne) mengandung asam lemak yang lebih bervariasi, yaitu asam tridesilat, asam palmitat, benzene, 1,4-BIS(Trimethylsilyl), dan N-Ethyl-1,3-Dithioisoindoline.

Microalgae Picochlorum sp. has high potential as a biodiesel and carotenoid producer. Walne is a synthetic fertilizer that is often used in microalgae culture which contains a high source of nutrients and is equipped with macronutrient and micronutrient requirements for microalgae growth and development. However, the high price of Walne fertilizer makes microalgae cultivation very expensive. Microalgae can be cultivated heterotrophically by adding nutrients from organic waste to obtain abundant biomass and reduce production costs. One of the organic wastes that have the potential as additional nutrients in heterotrophic cultivation is Fermented Mother Liquor (FML). FML is a liquid waste from the manufacture of food seasoning which has a high crude protein and nitrogen content. The purpose of this study was to determine the dry weight of biomass, cell number, growth rate, fatty acid profile, lipid and carotenoid concentrations of Picochlorum sp. with variations in the composition of the FML and walne medium, namely P1 (0% FML + walne), P2 (0.01% FML + walne), P3 (0.01% FML), P4 (0.03% FML + walne), P5 (0.03% FML), P6 (0.06% FML + walne), and P7 (0.06% FML). This study used a one-factor Completely Randomized Design (CRD) with 4 repetitions. The parameters observed were biomass, cell number, lipid concentration, and carotenoids concentration which were analyzed quantitatively, while the fatty acid profile was analyzed descriptively qualitatively. The results showed that variations in the composition of FML and walne medium had an effect on dry weight of biomass, growth rate, lipid and carotenoid concentrations of Picochlorum sp. The highest biomass and growth rates were obtained at P6 (0.06% FML + walne) of 0.115 grams and 299 cells/day. The highest lipid concentration was obtained in the P2 treatment (0.01% FML + walne) of 29.62%. Meanwhile, the highest concentration of carotenoids was obtained in the P3 treatment (0.01% FML) of 36.104 μg/mL. The results of the fatty acid profile treatment P1 (0% FML + walne) contained more varied fatty acids, namely tridesylic acid, palmitic acid, benzene, 1,4-BIS(Trimethylsilyl), and N-Ethyl-1,3-Dithioisoindoline.
