Abstract
Penyakit hipertensi dan diabetes melitus masih menjadi tantangan kesehatan di Indonesia, sehingga diperlukan pemanfaatan tanaman obat seperti daun dewa (Gynura pseudochina) yang kaya akan metabolit sekunder, salah satunya adalah flavonoid. Peningkatan kandungan flavonoid dapat dilakukan melalui pendekatan elisitasi, yaitu pemberian senyawa tertentu untuk memicu jalur biosintesis metabolit sekunder pada tanaman. Polietilen glikol (PEG) 6000 merupakan senyawa osmotik non-penetran yang mampu merangsang jalur fenilpropanoid yang berperan dalam sintesis flavonoid, fenol, dan alkaloid. Penelitian ini bertujuan untuk mengetahui pengaruh elisitor polietilen glikol (PEG) 6000 terhadap respon pertumbuhan dan kandungan flavonoid total pada kultur in vitro daun dewa. Hasil penelitian menunjukkan bahwa perlakuan PEG 1% merupakan perlakuan optimal, ditandai dengan waktu muncul tunas (5,00 hari), luas daun (0,908 cm2), berat basah (0,48 g), dan kandungan flavonoid total (6,94 mgQE/g). Sebaliknya, perlakuan PEG 5% menunjukkan hasil kurang optimal, dengan penurunan persentase eksplan berakar menjadi 66,7 ± 0,52%, tinggi eksplan (3,61 cm), luas daun (0,085 cm2), berat basah (0,33 g), jumlah akar (4,25), panjang akar (2,69 cm), dan kandungan flavonoid total (6,32 mgQE/g). Secara keseluruhan, PEG 6000 pada konsentrasi 1% mampu meningkatkan pertumbuhan dan akumulasi flavonoid, sedangkan konsentrasi 5% memberikan efek penghambatan akibat kondisi osmotik yang diterima.

Hypertension and diabetes mellitus remain health challenges in Indonesia, highlighting the need for the utilization of medicinal plants such as Gynura pseudochina, which is rich in flavonoids. The enhancement of flavonoid content can be achieved through elicitation approaches. Polyethylene glycol (PEG) 6000 is a non-penetrating osmotic agent that can stimulate the phenylpropanoid pathway, which plays an important role in the synthesis of flavonoids, phenolics, and alkaloids. This study aimed to determine the effect of polyethylene glycol (PEG) 6000 as an elicitor on growth response and total flavonoid content in in vitro cultures of Gynura pseudochina. The results showed that the 1% PEG treatment was the most optimal, as indicated by the fastest shoot emergence (5,00 days), highest leaf area (0,908 cm2), highest fresh weight (0,48 g), and highest total flavonoid content (6,94 mgQE/g). In contrast, the 5% PEG treatment showed the poorest results, with a decrease in rooting percentage to 66,7 ± 0,52%, shoot height (3,61 cm), leaf area (0,085 cm2), fresh weight (0,33 g), root number (4,25), root length (2,69 cm), and total flavonoid content (6,32 mgQE/g). Overall, PEG concentration 1% enhanced both growth and flavonoid accumulation, while concentration 5% inhibited plant development due to excessive osmotic condition.
